| Objective A common feature of athletics is full of drastic conflictions and competitions.The ankle parenchyma of athletes may get hurt in certain sports because of one over-extent action or a long time intensive training,which is easy to cause arthropathy and will not heal in a short time. In some serious condition,it even causes deformity.Joint inflammation is the most important reason for joint injury. cronic pain is the primary symptoms of troubled athletes with Arthritis.At this stage of the study,we observate concentration changes of metalloproteinase-3(Matrix Metalloproteinase-3,MMP-3) in serum and inflammatory tumor necrosis factor-associated factor(TNF-α),IL-1β(IL-1β), IL-6(IL-6) as well as the neurotransmitterγ-aminobutyric acid(gamma-aminobutyric acid,GABA) mRNA expression levels in lumbar spinal cord.To Ibuprofen for the positive control,we investigated the possible molecular mechanism of inflammatory joint pain resulted in sports injury.Methods:Results:64 grown-up male SD rats,which were randonly divided into the sham-ooperation group,the physiological saline group,the HWTX-I group and ibuprofen group.We induced rat models of adjuvant arthritis combined with Yaksh T.L,Rudy T. A catheterization in spinal epidural space in the study, relative anti-inflammatory drugs were given to rats through the reserved PE10 plastic tubes or intragastric administration. RT-PCR was used to detect the level of TNF-α,IL-1β,IL-6, GABA expression in lumbar spinal cord,and ELISA was used to detect the concentration of MMP-3 in serum.Results:(1) The result of pain test of joints injured side shown that, HWTX-I epidural injection drug use as well as the gastrointestinal tract ibuprofen can significantly reduce the inflammatory injury of bone and joint injury in rats lateral joint pain behavioral response(2) The result of circumference measurement of joints injured side shown that,HWTX-I as well as ibuprofen had obvious anti-arthredema efficacy after epidurally administered.(3) The results of ELISA shown that concentration of MMP-3 in serum in the non-treatment group was significantly higher than the HWTX-I treatment group(P<0.05) and ibuprofen treatment group(P<0.05).(4) RT-PCR have shown that TNF-α,IL-1β,IL-6 mRNA expressed in physiological saline group are much more higher than ibuprofen group and HWTX-I group(P<0.01).(5) RT-PCR have shown thatGABA mRNA expressed in HWTX-I group is lower than ibuprofen group(P<0.05) and physiological saline group(P<0.01).Conclusion:(1) In our research,we evaluated the ethology of rats with pain test,the result showed that HWTX-I and ibuprofen had obvious analgesia efficacy compared with physiological saline.(2) The ankle joint circumferences have been measured,the result showed that HWTX-I and ibuprofen could eliminate arthrocele in some degree.(3) HWTX-I injected epidural as well as ibuprofen which filled the gastrointestinal tract can reduce concentrations of MMP-3 in serum of rats with inflammatory bone and joint injury,and its possible molecular mechanism is that the two drugs arthritis reduced the symptoms of arthritis and cartilage destruction and achieved the purpose of pain relief by inhibiting over-expression of MMP-3.(4) HWTX-I injected epidural as well as ibuprofen which filled the gastrointestinal tract can reduce the expression of mRNA of inflammatory cytokines(TNF-α,IL—1β,IL-6) and neurotransmitter(GABA) in lumbar spinal cord of rats with inflammatory bone and joint injury,which showed that emergency of inflammatory-pain related with the release of inflammatory-activated neurotransmitter. |
