Objective: chronic atrophic gastritis is an intractable frequently-occurring disease of digestive system. It is characterized by gastric mucosal atrophy thinning, glands reduced or disappeared.Traditional chinese medicine attributed it to "Epigastralgia", "Lump in the abdomen or above" and so on. Modern medicine to the cause of CAG have not yet been fully clarified, the treatment also lacks effective methods. Traditional chinese medicine in the treatment of the disease shows unique advantages,with syndrome differentiation, flexible addition and subtraction, small side effects.After many years of clinical, teaching, scientific research study, at the conclusion of previous theory and experience, combined with changes of today's lifestyle, diet, etc. Instructor thinks that " Toxic cloud" intrinsic is the key to pathogenesis of the CAG, and tries to use HuaZhuoJieDuFang (capillaris, huanglian, angelica, paeony, danshen, qingbanxia, agastache, hedyotis diffusa, scutellaria barbata, villosum, notoginseng powder) in the treatment of CAG. Its treatment has opened up a new idea, for more than four decades of clinical application and efficacy is satisfied. In order to further explore this effect and mechanism, we carried out an experimental study on animals. General method is used in this experiment and to create CAG rat model.Using normal rats and CAG rats as research subjects and Victoria Su-enzyme as a positive control drugs, by observing the impact of HuaZhuoJieDuFang on CAG rats about normal situation,gastric pathology, epidermal growth factor (EGF), the gastrin (GAS), prostaglandinE2(PGE2), to explore the mechanism of CAG for HuazhuoJieduFang and provide a theoretical basis for clinical application.Method: Feeding Wistar 60 rats one week adaptatively, then in accordance with random number table , 10 randomly selected as the normal group, and the remaining are model.In the process of this10 rats were dead(four in them were killed by obersoering the effect of model).After model were successfully created , the remaining 40 were randomly divided into 4 groups:model group, Victoria Su-enzyme group, HuaZhuoJieDuFang high-dose group (referred to as high-dose group), HuaZhuoJieDuFang low-dose group (referred to as low-dose group), 10 each group. In addition to the normal group, the remaining rats in each group are 2% sodium salicylate and 30% ethanol mixed solution gavage, 0.1% ammonia water to drink freely combined with hunger satiety disorders, 90 days later the models were created successfully.After the rats model of CAG is successful,treatment was given the next day:the normal control group and model group were given equal volume saline, control group were fed with Victoria Su-enzyme suspension which density is 0.1g / ml, 10ml/kg/d, high-dose group were fed with HuaZhuoJieDuFang decoction, which including 71.4g pharmacognosy /kg/d, low-dose group were fed with HuaZhuoJieDuFangdecoction, which including 23.8g pharmacognosy /kg/d. Each group was administered for 60 days. At the end of fasting times after administration, cut-water 24 hours, decapitated blood, each 4ml, to do the following indicators: epidermal growth factor (EGF),gastrin (GAS) ,and collecting specimens of gastric determination to do the following indicators: Gross observation of gastric mucosa, gastric mucosa pathological observation, prostaglandinE2 (PGE2).Results: (1) gastric general observation: by observing folds, mucous membrane color,and bleeding,we found that the model group plica reduced or disappeared, less mucus, mucous membrane color whitish or gray, and some are accompanied by scattered bleeding points. After Victoria Su-enzyme,low-dose and high-dose HuaZhuoJieDuFang therapy, gastric mucosal damage are in varying degrees of improvement to the most obvious high-dose group. (2) Gastric mucosa pathology observation: by observing the quantity of gastric mucosa intrinsic glands, exit inflammatory cell infiltration or not etc.,in the model group significantly reduced the number of inherent glands, with disorder, and inflammatory cell infiltration. After Victoria Su-enzyme,low-dose herb and high-dose herb therapy, gastric atrophy are varying degrees of reversal to the most obvious high-dose group. (3) the impact on epidermal growth factor (EGF): In model group serum EGF was significantly higher than the normal group, the two have significant differences in comparison (P<0.001); HuaZhuoJieDuFang high-dose and low-dose group and Victoria Su-enzyme group can reduce serum EGF content significantly, comparing to model group there is significant difference (P<0.001),it shows that HuaZhuoJieDuFang and Victoria Su-enzyme can control serum EGF content effectively; In the three treating groups serum EGF concen is close to normal,comparing with it there is no significant differences (P>0.05). In three treating group , serum EGF content was no significant difference (P>0.05) among them, but the effect of high-dose group the most obvious.(4) the impact of serum gastrin (GAS): compared with normal, serum GAS of model group were significantly lower there is significant difference between them (P<0.001). After Victoria Su-enzyme and low-dose HuaZhuo JieDuFang are used,serum GAS content increases,but comparing to normal group it have obvious significance(P<0.01),but between them there is no significant difference (P>0.05). High-dose can increase serum GAS level,comparing with the model group there has significant difference (P<0.001);comparing with Victoria Su-enzyme and low-dose group,there has diference among them(P<0.05).It shows that high-dose HuaZhuoJieDuFang may adjust the level of gastrointestinal hormones so as to protect the gastric mucosa.(5) the impact of prostaglandin E2 (PGE2): Compared with normal group ,PGE2 content in gastric mucosa in model group decreased significantly, it has significance (P <0.001), compared Victoria Su- enzyme and low-dose group to the normal group ,there has significantly different (P<0.001 ), but both them can increase PGE2 content. Compared model group with high-does and low-dose group and the Victoria Su-enzyme group PGE2 content in gastric mucosa increased significantly, there is significance among them (P<0.001, P <0.05, P<0.01), Victoria Su- enzyme group and low-dose group are less effective than high-dose group, there has significant difference (P<0.001),but between them there is no significant difference(P>0.05).It shows that high-dose HuaZhuoJieDuFang is better than another groups on adjusting gastric mucosa protection factor.Conclusion: This experiment proved that HuazhuoJieDu Fang have the following effects: (1) to improve the morphology of gastric mucosa histopathology; (2) to control the expression level of EGF for the status improvmente of gastric mucosa and gastric mucosal repairation. (3)to promote GAS secretion, stimulate gastric acid secretion, atrophic glands reversation; (4)to promote the release of PGE2, and enhance its protection role of gastric mucosa.Its mechanism are possible by adjusting the level of gastrointestinal hormones ,increasing gastric mucosal blood flow, improving microcirculation and enhancing mucosal barrier defensing and repairing functions, so as to achieve for the treatment of chronic atrophic gastritis purpose.
