Electroacupuncture is beneficial to the degestive system which has been comfirmed by a vast of reseaches. While its mechanism is still unknown, So the further interpretation about the mechanism of electroacupuncture-regulated gastric motility is very important to guide clinical treatment. This intestigation observed the interstitial cells of cajal(ICCs), connexin 43(CX43) and extracellular signal regulatedkinase(ERK) which are connect to ICCs by electroacupuncture on the Tsusanli on the basis of electrogastrogram. GeneChip technique was used to study the variation of correlated difference genes and signal transduction pathways of electroacupuncture-regulated gastric motility on rat's Tsusanli synchronously. The new theory will be provided to evaluate the possible mechanism of electroacupuncture's regulatory effect on gastric motility function.Methods:The rats were randomized divided into three groups, The first group with electric acupuncture on the Tsusanli (ST36,Tsusanli group), the second group with electric acupuncture on non-specific point (the non-acupoint group), the third group is control with non-acupuncture. Electrophysiological method was used to observe the changes of gastric electrical activity respectively. We observed distribution of ICCs-CX43 and ICCs-ERK in gastic body by immuno-fluorescence double-stain of immunohistochemistry assay. GeneChip technique was used. We isolated the Total RNA from gastric tissue samples of experimental rats by the TRIZOL method. And the cRNA probe with biotin-labeling was obtained by reverse transcription method. Then the cRNA probe was hybridized with Oligo GEArray GeneChip(American SuperArray Corporation). Fluorescence signals of gene array were scanned and the differences of the genes expression and the signal transduction pathways among the three groups were analyzed by computer.Results:1. Electroacupuncture can affect the gastric electroactivity obviously┄increasing its amplitude and frequency. It suggested that electroacupuncture at Tsusanli can promote the gastric motility significantly.2. The expression of CX43 and ERK which is connected with ICCs of Tsusanli group in myenteron and muscular layer increased obviously.3. Correlated difference genes and signal transduction pathways of electroacupuncture have been selected. The marker genes expressed in three groups. In Tsusanli group, Hoxa1, lep, bcl-2 genes were up regulated, which participates in retinoic acid pathway, insulin pathway, survival pathway, estrogen pathway and phospholipase C pathway respectively, as well as Ptgs2 gene was down regulated which participates in phospholipase C pathway. The up regulation of lep and the down regulation of Ptgs2 were correlated to ICC's activation. It may produce a marked effect in gastric motility function.Conclusions:Electroacupuncture at Tsusanli can promote the gastric motility obviously. The effect of electroacupuncture at Tsusanli is carried out mainly through promoting the electroactivity of ICCs. Electroacupuncture at Tsusanli can increase the expression of CX43 and ERK which are closely to ICCs. The mechanism of electroacupuncture-regulated gastric motility in rats is related to the correlated genes, which are up or down regulated. Moreover, it is also related to some specific signal transduction pathways.
