ObjectiveTo research the influence of leech extract on human retinal pigment epithelial(hRPE) cell's signal transduction that is guided by P38 mitogen activated protein kinase.Methods(1) The primary human retinal pigment epithelial cell that was purchased was anabiosised and then cultured in vitro.(2) Based on the previous experiments, 64mg/ml leech extract and 0.5NIHU/ml thrombin were used to incubation hRPE 30minutes.The expression of P38MAPK was measured by flow cytometry and immunocytochemistry.Results(1)The fluorescence expression of P38MAPK display: thrombin group(6.4417±0.072) compared with normal control group(5.4567±0.1825),the fluorescence quantity increased(P<0.01);leech extract group(4.4483±0.2015) compared with normal control group,the fluorescence quantity decreased(P<0.01);added leech extract and thrombin at equal pace(4.1433±0.068) compared with thrombin group, the fluorescence quantity decreased(P<0.01).(2)The optical density (OD) value of P38MAPK display: thrombin group (1.195±0.0226) compared with normal control group(1.055±0.0187),OD increased(P<0.01); leech extract group(0.7567±0.071) compared with normal control group,OD decreased(P<0.01);added leech extract and thrombin at equal pace(0.555±0.065) compared with thrombin group, OD decreased(P<0.01); ConclusionLeech extract can inhibit the proliferation of hRPE by P38 mitogen activeated protein kinase. |
