| As the worldwide prolongation of people's life expectancy and acceleration of the aging of population, nowadays cardiovascular disease has become one of the main reasons of adult death in China. Among all reasons of this adult death, cerebrovascular disease is the NO.1 cause of disability and second cause of death. To be exact, dyslipidemia is the root and most dangerous factor which result in cardiovascular disease's occurence and development. Therefore, the prevention and control of dyslipidemia has a great meaning in decreasing the rate of cardiovascular disease and improve its prognosis. At the same time, hyperlipidemia can increase the microviscosity of red cell membrane and decrease its transformation ability by effecting lipidal double layers and scaffolding protein of red cell membrane. Since red cell membrane turns to spiniform, the area of cell membrane grows, blood viscosity increases, all these lead to abnormal hemorheology, increased haematocrit, blood steam and microcirculation slow down, then it brings about the tissue short of blood anoxic and metabolic disturbance. Usually, hemorheology could changes before the emergence of clinical symptoms of cardiovascular disease, and it plays an important role in the early discovery and treatment of it.Generally speaking, lipid-lowering drugs can be induced into 5 ways by its effects and mechanism. 1. Prevent absorbability of cholic acid or cholesterin from intestinal canal and improve the discharging of them together with excrement; 2. Restrain composition of cholesterin in the body, or promote transformation of cholesterin; 3.Promote expression of LDL's receptor on cell membrane and accelerate decomposition of lipoprotein; 4.Activate lipoprotein to metabolize enzymes and stimulate the hydrolyzation of triglyceride; 5. Prevent the in-body composition of other adipose fin or promote the metabolism of them. There are many kinds of lipid-lowering drugs, and they can be divided into 4 types by their functions, decrease CHO, mainly decrease CHO and simultaneously decrease triglyceride, decrease triglyceride, mainly decrease triglyceride and simultaneously decrease CHO.Although western medicine is definitely curative in regulating blood lipid, it also has different side effects to some extent. At present, people accepted Statins as the most effective western medicine and it is widely used to regulate blood lipid, however, it is very expensive. What's more, such side effects which toxicity to liver and muscle and so on even could cause rhabdomyolysis seriously. For instance, owing to its side effects, lipid-lowering drug Baycol has been forbidden to be used. Using traditional Chinese medicine to regulate blood lipid becomes a hot research field for its superior curative effects and few side effects. Looking on the holism concept, traditional Chinese medicine can intervene in the formation and treatment of hyperlipidemia in many processes.This experiment took rats with hyperlipidemia as research object to observe how Ju Teng Capsule affected their blood lipid, hemorheology and discuss the capsule's effects and mechanism to molecule level.Experimentâ… Ju Teng Capsule's effects on Lipids and High DensityLipoprotein Receptor Liver Gene Expression in hyperlipidemia rats.AIM: To study the regulatory mechanism of lipid metabolism disorders in the blood fat of hyperlipemia rat model with Ju Teng Capsule, and do systematic observation on the functions of this medicine on lipids and high density lipoprotein receptor in rat liver gene expression, and then to clarify the mechanism of action of this medicine on clinical application. METHODS: Selected 60 SD rats as investigated subject, and divided them into six groups randomly: group A, group B, group C, group D, group E, and group F. Each group included ten rats. Group A was normal group, group B was model group, group C was Xue Zhi Kang group, and group D, E and F were Ju Teng Capsule groups with low, medium and high dosage respectively. Croup A was fed normal forage, but other groups were fed high fat forage. All groups were allowed to drink water freely. Group A was fed normal forage without intragastric administration; group B was received physiological saline 10 ml/kg·d with intragastric administration; group C was received Xue Zhi Kang Capsule 0.090g/ kg·d with intragastric administration; group D, E and F received Ju Teng Capsule 1.175g / kg·d, 2.350g / kg·d, 4.700g / kg·d with intragastric administration respectively; administered for six weeks continuously. Then the samples in the research were collected and examined the level of TC, TG HDL-C and LDL-C in the blood serum; analyzed the changes of SR-BI gene expression in rat's liver by RT-PCR and Western blot. RESULTS: Ju Teng Capsule can markedly decreased the level of TC, TG, LDC-C in the blood serum, raise the level of HDL-C, enhanced SR-BI gene expression in rats'liver. Results: Compared with the normal group, the plasma viscosity, low, medium, high share rate, red cell deformation index, haematocrit, stagnation of red cell, fibrinogen in plasma and MPAG of model group rats were significantly different (P<0.05). Ju Teng Capsule groups notably differed with the model group in reducing plasma viscosity and whole blood viscosity of medium, high share rate (P<0.05). Although the whole blood viscosity of low share rate trended to decrease, it had no statistics meaning (P>0.05). Ju Teng Capsule could obviously increase red cell deformation index, decrease haematocrit and aggregation index of red cell (P<0.05), and the packing fraction of fibrinogen in plasma, MPAG (P<0.05).Main conclusions:1. Ju Teng Capsule has an applicable value on preventing the hyperlipidemia, enhancing SR-BI gene expression in rats'liver and finally accomplishing the purpose of reducing blood fat and development of hyperlipidemia and its complications.2. Ju Teng capsule could obviously improve hemorheology in hyperlipedemia rats, reduce blood viscosity of them, improve the spissitude, viscous and congeal, gathering states of blood. Its mechanism to decrease blood viscosity can be divided into following aspects: decrease hematokrit, decrease contents of fibrinogen, restrain aggregation of blood platelet, and enhance deformability of red cell. |
