Effects And Molecular Mechanisms Of Matrine On Human Hepatocarcinoma SMMC-7721 Cells

Objective 1.To investigate the refrain role of proliferation on SMMC-7721 cells acted by Matrine.2.To find out the Livin,Survivin gene in the process of apoptosis.Methods Effects of Matrine on SMMC-7721 cell proliferation was analyzed by MTS assay;Cell apoptosis were examined by flow cytometry(FCM);RT-PCR was used to observe the expression of Livin/Survivin genes and Western was used to observe the expression of Livin/Survivin protein.Results 1.Compared to control group,Matrine can obviously refrain proliferation of SMMC-7721 cells(P＜0.05);2.After treated with matrine for 48 hours,compared to control group,cell apoptosis increased significantly,and this effect of matrine was in a dose-dependent manner,3.After treated with matrine for 48 hours,RT-PCR and Western showed the expression of Livin/Survivin decreased.Conclusion Matrine can strongly refrain the growth of SMMC-7721 Human Hepatocarcinoma SMMC-7721 cells,Matrine can induce the apoptosis of SMMC-7721 cells,Livin and Survivin are very important apoptosis regulation gene,and play a role in the apoptosis of SMMC-7721 cells treated by matrine.
