Objective:To investigate the effect of the proliferation and apoptosis of Baicalein on hepatocellular carcinoma cell line SMMC-7721 in vitro.The purpose of this study were provided more and sufficient theoretical evidence for Baicalein in hepatocellular carcinoma clinical treatment.Methods:The cells exponential phase of growth were made into cells suspension,2.5×104 cells per well were inoculated into 96-well culture plates. After 6h-12h, Baicalein solut was added into each well in which the final concentration of Baicalein were 50μmol/L, 100μmol/L,150μmol/L,200μmol/L. After 24h、48h, the cell line proliferation was assayed by using MTT. Absorbance value (A) was detected and inhibitor ratio was calculated by the formulation:IR=[1-(A in experimental group-A in blank group)/(A in control group-A in blank group)]×100%.2. The cells in exponential phase of growth were made into cell suspension,3.0×106 cells per well were inoculated into 6-well culture plates. After cell attachment, Baicalein solut was added into each well in which the final concentration of Baicalein were 50μmol/L,100μmol/L,150μmol/L. After 48h, The effect of the apoptosis of Baicalein on SMMC-7721 cells was detected using flow cytometry.3. The cells in exponential phase of growth were made into cell suspension,2×105 cells per well were inoculated into 6-well culture plates. After cell attachment, Baicalein solut was added into each well in which the final concentration of Baicalein were 50μmol/L,100μmol/L. After 48h, the mRNA levels of Survivin and the mutation of p53 in SMMC-7721 were analyzed by reverse transcription polymerase reaction(RT-PCR).4. All the experimental results were analyzed by SPSS19.0. Results:1. The proliferation of SMMC-7721 cell line in the experimental group was singnificantly inhibited compared with the control group(P<0.05).2. The apoptosis rate of SMMC-7721 cell line in the experimental group was singnificantly raised compared with the control group(P<0.05).3. The mRNA expression levels of Survivin and the mutation of p53 in SMMC-721 cell line of the experimental group were singnificantly down-regulated compared with the control group(P<0.05). Conclusions: 1.Baicalein may inhibit the proliferation of SMMC-7721 cell line.2. Baicalein may induce apoptosis of SMMC-7721 cell.3.Baicalein may down-regulate the expressions of Survivin and the mutation of p53. |
