Objection:To investigate the effect of small interference RNA targeting polo-like kinase 1 on inducing cells proliferation and apoptosis in human lung carcinoma cell line A549 cells.Methods:The liposome(Lipofectamine 2000) containing different doses of siRNA(Small interference RNA)targeting PLK1 gene were transfected into A549 cells.The transfection ratio was evaluated by observation under fluorescent microscopy.The cell proliferation and apoptosis was evaluated by TUNEL assay.Results:After the transfection,We found the differences between four different groups,the proliferation ratio was found decrease and apoptosis ratio increase,due to the rising doses of siRNA,through TUNEL assay.The apoptosis in the group of 50nM was significant increased compared to the other groups.There was no significant difference in the effects of proliferation and apoptosis between the 48 and 72 hours after tansfection. Conclusions:PLK1 targeting siRNA could inhibit the expression of PLK1 gene and cell proliferation,as well as inducing cells apoptosis in human lung carcinoma cell line A549 cells.
