The Population Pharmacokinetics Research Of Flushing Dose MTX For Acute Lymphoblastic Leukemia In Child Endosomatic

ObjectiveTo evaluate the Population pharmacokinetics of high-dose methotrexate in children with acute lymphoblastic leukemia though monitoring the plasma concentration,which can offer necessary indicatrix for adjusting individual dosage regimen.MethodsCollected the MTX plasma concentration of the 32 children(16male and 16female)with ALL who were in our hospital from 2006 to 2008.Population pharmacokinetics and the variation of intersubject and intrasubject were measured by NONMEM software.and evaluated the fix effect lector such as age,gender,weight, ALT.AST,GREA on Population pharmacokinetics parameters and obtained the final regression modle.ResultsThe parameter of PPK is CL=7.09(L·h^-1),V=30.2(L),Kα=0.5.ALT,AST and Gender has statistic influence on CL.ConlusionThis study established the detabase about the therapy of high-dose MTX in children with acute lymphoblastic leukemia,Population pharmacokinetics model equtation were made through NONMEM,and Get Population pharmacokinetics parameters which can offer important indicatrix for MTX clinical individualized treatment.
