ObjectivesFor a long period,atherosclerosis(As) has been regarded as endothelial inflammatory disease,and the function of vascular adventitia is just nourishing and supporting vessels.However,recent researches have found that vascular adventitia plays an important role in the regulation of vascular function.The sympathetics and parasympathetics nerve terminal in vascular adventitia release neurotransmitters.These substances act on medial smooth muscle cells and adjust vasomotor function.The main components of vascular adventitia,fibroblast cell,can be activated in pathological conditions.Then the fibroblast cells may have phenotype conversion,proliferation, migration and synthesis of a variety of cytokines and take part in the development of atherosclerosis.It has been confirmed that the adventitial inflammation exists in blood vessels of As.The infiltration of inflammatory cells in vascular adventitia has a positive correlation with vascular membrane lesions of As.Some reports show that adventitial inflammation has some relations with endothelial dysfunction and the occurrence of acute coronary syndrome.The vasa vasorum of blood vessel may be the gateway of the inflammatory cells invasion.However,the mechanism of adventitial inflammation of As is not yet clear.The researches are rarely in the world that about the role of the adventitial inflammation and whether TongXinLuo can inhibit adventitial inflammation and improve artery endothelial function.Thus,through a series studies of animal experiments,we aim to observe the relationship between artery adventitial inflammation and endothelial function as well as the interventional function of TongXinLuo.Materials and Methods1.Establishment of rat right common carotid artery(RCA) injury model and experimental grouping:Thirty male Wistar rats weighing 200~250g were randomly divided into 3 groups:the normal group,model group and Tong Xin Luo treat group. After operation,The rats were fed standard food and free water for7 days.2.The rats right common carotid artery were taken and were stained with HE method to observe artery adventitial inflammation,media and intimal hyperplasia by light microscopy.3.All rats were killed in each group on 7 th day after operation and the blood 5 ml was sampled from the heart for use before killing.Determination of the serum level of nitric oxide(NO),nitric oxide synthase(NOS) and endothelin(ET) concentration was made respectively by nitric reductase and ELISA.4.Statistical analysis:All the data were analyzed using SPSS13.0 software package for statistical analysis.Data were expressed as the mean±standard deviation ((?)±s).Group comparison was done using analysis of One-Way ANOVA.AP-value of less than 0.05(p<0.05) was considered to be statistically significant Result.Results1.Pathological and morphological changes of the rat right common carotid artery specimen.In the normal group,we didn't observe artery adventifial inflammation and the artery intima in rats remains intact by light microscopy.The artery adventitial inflammation,media thickening and the artery intimal inflammation in rats existed on the 7th day in the arteries of model group.But it was not significant in TongXinLuo group.2.Serum NO,NOS and plasma ET levelsCompared with model group,the serum levels of NO and NOS in TongXinLuo group were higher(p<0.05);but the level of ET in TongXinLuo group was lower than that in model group(p<0.05).Conclusions1.The artery adventitial inflammation took part in atherosclerosis.2.The artery adventitial inflammation took part in the pathogenesis of endothelial dysfunction in the collared femoral artery of the rat.3.TongXinLuo can inhibit adventitial inflammation and improve artery endothelial function by decreasing the plasma level of ET and increasing the serum levels NO and NOS in the collared artery.
