ObjectivesAtherosclerosis (Atherosclerosis, AS) is an inflammatory disease rather than simply due to the result of lipid deposition is the result of many factors, many inflammatory cytokines involved in the formation of AS. In which the immune response of IL as indicators of inflammation, the elevated levels were significantly associated with cardiovascular events. Recent research confirms that vascular adventitial injury in some individuals could be the origin AS start portal. To this end the establishment of the present study, cuff injury model of rat carotid artery adventitia and ultrafine powder for TXL (TXL) intervention, membrane injury of vascular cell signal transduction pathways-Nuclear transcription factor-κB (NF-κB) and serum interleukin-6 (IL-6), interleukin-10 (IL-10) levels, to explore the outer membrane and the formation of inflammation in atherosclerosis and the mechanism of Tongxinluo Protective effects of ultrafine powder.Materials and Methods1. Establishment of rat left common carotid artery (LCA) injury model and experimental grouping:Thirty male Wistar rats weighing 200-270g were randomly divided into 3 groups:the control group, model group and Tong Xin Luo treat group. After operation, The rats were fed standard food and free water for7 days.2. The rats left common carotid artery were taken and were stained with immunohistochemical method to observe and counting artery wall activation of NF-κB positive.3. All rats were killed in each group on 7th day after operation and the blood 5 ml was sampled from the heart for use before killing. Determination of the serum level of IL-6,IL-10 concentration was made respectively by ELISA.4. Statistical analysis:All the data were analyzed using SPSS16.0 software package for statistical analysis. Data were expressed as the mean±tandard deviation (x±s). Group comparison was done using analysis of One-Way ANOVA. Non-parametric data groups were analyzed using Kruskal-Wallis H analysis of rank sum test between the two groups using Mann-Whitney U rank sum test analysis. AP-value of less than 0.05(p<0.05) was considered to be statistically significant Result.Results1. Vascular tissue immunohistochemistry resultsLight microscope:nuclear factor-KB p65 normally brownish yellow granules, the nucleus is almost no staining; activated to enter the nucleus, showed nuclear staining. Almost no nuclear translocation of the control group; Vascular adventitial injury group 7 days after the vessel wall cells can be seen more brown nuclei; TXL treatment group saw little brown nucleus, significantly less than the outer membrane group NF-icBp65 membrane group was significantly stronger than the control group, the difference was statistically significant (u=3.67, p<0.05); NF-κBp65 expression TXL group compared with the outer membrane decreased, the difference was statistically significant (u=2.83, p<0.05); TXL Group expression compared with the control group there was nonsignificant difference (u=1.77<1.96, p>0.05).2. Serum IL-6, IL-10 determination resultsCompared with the control group, serum adventitia IL-6, IL-10 levels were increased (P<0.05); with the outer membrane group, TXL serum levels of IL-6, IL-10 levels were reduced (P<0.05), compared with the control group, TXL Group also increased between the serum concentration, but did not achieve statistical significance (P>0.05).Conclusion 1. Vessels the outer membrane of the casing of atherosclerosis great success and built a model, which mechanisms and inflammatory factor NF-κB, IL-6, IL-10 related.2. Tongxinluo can reduce inflammation and atherosclerosis lesions.
