Studies On The Scanning Of Chinese Crude Drug Using For Inhibiting β-hemolytic Streptococcus And Removing-immune Complex

Acute glomerulonephritis usually occurred after be affected by beta-hemolytic streptococci.M protein or other ingredient(antigen) that was be found in the cell wall of streptococcus can induce organism to product antibody,the formative immune complex may sediment in glomerular basement membrane,and it may caused immunopathogenesis injury.For exploring Chinese traditional medicines which have better curative effect that was use for curing acute glomerulonephritis.This paper carried out a bacteriostasis trial to beta-hemolytic streptococci by 16 species Chinese herbal medicines,and comparative experiment about the effects of removing immune complex by 5 species Chinese traditional medicines and 3 species polysaccharide.The Chinese herbal medicines that are included(Chinese Magnolivine Fruit,Szechwan Lovage Rhizome, Tangshen,Tangshen,Isatis Root,Honeysuckle Flower,Climbing groundsel,Radix Glycyrrhizae,Rhubarb, Peppermint,Skullcap Root,Ginkgo leaf,Giant Knotweed Rhizome,Amur Cork-tree,Golden Thread and Propolis) contained substance with inhibitive activity such as(organic acid,alkaloid,ftavone,volatile oil, quinone) were selected in this paper.We took different conditions extracting these medicines and used double liquid dilute method for the determination of minimum inhibition concentration.The extracting conditions were:water,50%alcohol,75%alcohol,95%alcohol.And we determined the inhibitive effects by paper spreading method.We got the effects of the same Chinese herbal medicine in various extracting conditions, and compared the anti-bacteria ability of these extracting substances.Water extracting substance with the best inhibitive effects was liquorice,inhibition zone diameter was 25.6mm.Water extracting substance of ginkgo leaf,Isatis Root,propolis and Climbing groundsel's inhibitive effects were not obvious.50%alcohol and 75% alcohol extracting substance with the best inhibitive effects was also liquorice.50%alcohol extracting substance of Baical Skullcap Root and ginkgo leaf's inhibitive effects were not obvious.All of 95%alcohol extracting substance have obvious inhibitive effects,and the best one was Chinese Magnolivine Fruit.We chose three Chinese herbal medicines with better inhibition effects from water extraction,50% alcohol extraction,75%alcohol extraction and 95%alcohol extraction respectively,then determined their joint inhibition effects using the method of chessboard serial dilution.The water extracting substance of Golden Thread and Chinese Magnolivine Fruit have addition effect,50%alcohol extracting substance of Radix Glycyrrhizae and Szechwan Lovage Rhizome have addition effect,75%alcohol extracting substance of Golden thread and Rhubarb can reach synergistic effect,when they were combined,the lowest anti-bacteria concentration were 0.12mg/ml and 0.24mg/mL.95%alcohol extracting substance of Rhubarb and Chinese Magnolivine fruit have addition effect,the lowest anti-bacteria concentration were 0.49mg/mL and 0.12mg/mL.At the same time,we got lentinan,ganoderan and Cordyceps sinensis polysaccharide and effective components of Safflower,Cortex moutan,Rehmannia dride rhizome,Peach Seed and Szechwan Lovage Rhizome through physical,chemical methods.We built nephritis model of animal and determined the content of immune complex in the(normal group,model group,injected group's) blood.Then we did the determinations of RBC-C₃b garlanding,immune complex garland and erythrocyte of immunizing regulatory factor.The conclusion were:szechwan lovage rhizome and ganoderan were the perfect medicines when enhancing the immune ability of erythrocyte.They are helpful for clearing immune complex,and they can promote the activity of RFER and reduce the activity of RFIR.For the clearing of immune complex that were in the circulation of blood and decreasing of immune complex to deposit in renal corpuscle basement membrane they have the best effects in this experiment.
