Objectives:To study the antibacterial and bactericidal ability of the traditional Chinese medicine,and to study the antibacterial mechanism of the traditional Chinese medicine and its monomer with good antibacterial effect.Methods:1.Select the effective 53 flavors of Chinese medicine with antibacterial effect,preparation of the decoction,sterilization standby.The establishment of high schoollow medicine decoction group and penicillin group and blank control group.The Oxford cup diffusion method and minimum inhibitory concentration(MIC)method for screening the antibacterial medicine has good effect on hemolytic streptococcus.2.For further evaluation of traditional Chinese medicine screening antibacterial ability,the cell membrane permeability was evaluated to determine the effect of bacterial virulence,and bacterial mortality were used.At the same time,penicillin group and blank group as control set.3.Selected 1 kinds of traditional Chinese medicine and its active ingredient of effective monomer,bacterial resting state,total RNA was extracted and sequenced,penicillin and normal saline is set as the control group.The results of sequence analysis,understand the differentially expressed genes of bacteria under different treatments.Make a preliminary study on the antibacterial mechanism of Chinese medicine from the level of gene transcription.Result:The bacteriostatic ring of 53 kinds of Chinese medicine and its related study of monomer on hemolytic streptococcus,minimum inhibitory concentration,cell membrane permeability,bacterial virulence and mortality of selected Lobelia chinensis and its monomer on the luteolin,hemolytic streptococcus gene expression.Conclusions:then get some conclusions below:1.53 herbs and active ingredients in Radix rehmanniae,Radix Scutellariae,Radix Scrophulariae,Rhizoma Atractylodis,lobelia,aster,Pyrrosia,Magnolia,aspidium,had better effect of luteolin and baicalin.2.The effects of traditional Chinese medicine and monomer on the virulence,cell membrane permeability and resting cell death rate of Streptococcus mutans were studied3.According to the transcriptome sequencing results,this study selected a total of 1046 differentially expressed genes.Compared with the control group,lobelia under the action of hemolytic streptococcus in cells responsible for bacteriocin,multi drug resistant MFS transport system,UDP-N-transposase,n-acetylmuramyl five amino acid N(6)-C aminoacyl transferase,lipoic acid salt and protein ligase a,ComX protein,B two aldehyde enzyme,PTS mannose transport auxiliary protein ManO ATP binding transporter protein,oxidoreductase gene expression increased significantly.Responsible for the penicillin binding protein 1A,uridine three-phosphate glucose-1-phosphate uridylyl transferase,ABC glucose transporter permease,5(benzyloxycarbonyl amino acid)imidazole nucleotide synthetase two,nicotinamide adenine nucleoside phosphate,expression of cysteine protease gene significantly decreased.Compared with the control group,luteolin group was significantly upregulated in the expression of the putative protein gene of Streptococcus,streptococcus is responsible for significant downregulation of putative protein 50S,ribosomal protein L21,expression of butterfly acid synthase two gene hydrogen.Compared with the control group,the penicillin group can significantly increase for Streptococcus hemolysin S gene significantly decreased responsible for uridine three phosphoric acid and glucose-1-phosphate uridylyl transferase gene.4.The results of differential expression gene GO function analysis showed that the genetic difference between Lobelia group and the saline control group,mainly involved in the RNA binding"," molecular structure activity "biosynthesis process","carbohydrate metabolism","lipid metabolism process","translation","molecular function of methyl transferase activity." gene differences in luteolin group and the saline control group is mainly involved in the alkyl transferase,aryl transferase "function.5.The Pathway analysis showed that KEGG gene expression,gene difference of hemolytic streptococcus lobelia and saline control group mainly involved in "polyketides and terpenoid metabolism" and "translation".Penicillin and saline control group genes of beta hemolytic streptococcus in carbohydrate.
