Matrine is the isomer of lupanine, widely existing in the roots of kurarinone, beans and broad beans. It is the main active ingredients of kurarinone herbs. It can improve the characteristics of the human immunodeficiency with little toxic side effects. It is always used as bactericidal anti-inflammatory, and is the main active ingredients of anti-cancer kushen drugs. Matrine is one of natural plant pesticides with high value application. In recent years, with environmental consciousness improved, and drug-resistant pests caused by chemical pesticides, pesticide residues in agricultural products and environmental pollution problems highlighted, it is necessary develope inevitable green pesticide. Supercritical fluid extraction technology is an emerging extraction technology with fastly transfer rate, high selectivity, low operating temperature, solvent-free residues, environmental protection and energy conservation. It has good development prospects in the food, traditional Chinese medicine processing, traditional Chinese medicine, biological engineering and other fields.In order to guide the extraction process and for the evaluation of the effects of various factors, the quantitative of matrine is determined in gas chromatography method. The results showed that: matrine can be separated in 30 m×0.32 mm(id)×0.25μm quartz capillary column, using phthalate isoamyl as the internal standard. Linear correlation coefficient is 0.999 1, the recoveries is between 98.5% and 102.6%, RSD=1.7%. The method is simple, rapid, accurate and reproducible and applies to the determination ofmatrine.Based on the extraction rate of matrine inspection targets, we study the conditions ofextraction process matrine in supercritical fluid. First, through single factor experiments, we got that the mainly affects the extraction pressure is between 25 MPa and 35 MPa; the main extraction temperature is is between 50℃and 60℃; the maximum extraction rate is in 40 mesh size raw materials; the extraction time is between 2.0 h and 4.0 h; the carbon dioxide flow is between 40 kg/h and 45 kg/h. Secondly, we discussed the different cosolvent of acetone, methanol, ethanol, tween-80, and determined the suitable concentration, volume and the combination conditions. Finally, we found the impact order of affecting the extraction rate matrine is temperature>Cosolvent>pressure, and the best condition is extraction temperature 55℃, extraction pressure 35 MPa , volume ratio of for ethanol, tween-80, distilled water 12 :1:7, extraction time 3.0 h. The average extraction rate of matrine can reach 1.28 percent in this condition.The method of supercritical carbon dioxide extracted from kushen has many advantages, such as the extraction with high dissolved capacity, a faster transfer rate, a better performance and the flow of balance, a good adjustable and ease of control, and to maintain matrine biological activity. The extraction has favored pesticide in the green industries of matrine for avoiding environmental pollution, non-toxic pollution-free process engineering, achieving the cleaner production, so the technology has considerable economic and broad prospects.
