Abstract/Summary:
Objective:Allergic diseases have increased dramatically in prevalence over the past two decades in economically developed countries.There is a key initiator of allergic Inflammation may exist.The mechanism that underlies the induction of allergic inflammation is not very clear.To find out the sensitization ways of the allergic disease have very important meaning.In our study,we focused on the diverse characteristics and the clinical significance of serum specific IgE of egg and cow milk in infants with allergic disease.And we found out that,The positive rates of egg sIgE in the stage of<6-months was 44%,The infants in the stage of<6 months even can not to get in touch with egg.So,sensitization through the gastrointestinal tract can not explain this kind of state,we considered that there might a way to sensitize the infants through the skin.Recent studies shows that thymic stromal lymphopoietin(TSLP) is one of these factors that acts very early in physiopathology of allergic diseas.TSLP may be a potential important factor in the induction of allergic disease.So we build sensitized mice model through skin murine models through skin to investigate the mechanisms of TSLP and in allergic diseases.Methods:1,Build mice model by OVA through skin in the new born mice.2,Observe the appearance and the appearance after stimulate to evaluate the overall allergic status.3,Detect expressions of TSLP in lung,in thymus,spleen,intestine with OVA sensitization by Real-time PCR.Results:1.OVA induced mice group and control group skin appearance:The rate of inflammation in OVA induced mice group and control group skin appearance:OVA induced mice group skin inflammation rate is 20%(3/15);control group is 0%(0/15)2.Appearance after provocation:OVA induced mice group appearance after provocation:egg white provocation red swelling rate:of the skin 0%(0/5),egg antigen for human provocation red swelling rate of the skin:0%(0/5),NS provocation red swelling rate of the skin:0(0/5)Control group appearance after provocation:egg white provocation red swelling rate of the skin:0%(0/5),egg antigen for human provocation red swelling rate of the skin:0%(0/5),NS provocation red swelling rate of the skin:0(0/5)3.Histopathology of lungs(stained with H&E):1).Histological analysis demonstrated thickness of bronchial epithelium,peribronchiolar and perivascular inflammatory infiltrates in the lungs of OVA group.Inflammatory changes mentioned above are not present in the control group.2).Skin of sensitization location:the cuticulum thickening and inflammatory cell infiltration is obviously.Inflammatory changes mentioned above are not present in the control group.3).Intestine:Inflammatory changes are not present in the OVA group and control group.4.The mRNA expression level of TSLP in OVA induced mice group and control group1).the mRNA expression level of TSLP increased of skin in OVA group with that in the control group,but has no significant difference with that in the control group (P=0.083)2).the mRNA expression level of TSLP increased of lung in OVA group with that in the control group,but has no significant difference with that in the control group (P=0.224)3).the mRNA expression level of TSLP increased of thymus in OVA group with that in the control group,but has no significant difference with that in the control group (P=0.277)4).the mRNA expression level of TSLP decreased of spleen in OVA group with that in the control group,but has no significant difference with that in the control group (P=0.217)5).the mRNA expression level of TSLP increased of intestine in OVA group with that in the control group,but has no significant difference with that in the control group(P=0.647)Conclusions1.New born mice show kind of allergic disease appearance by though skin sensitization.2.There is a increase tendency TSLP expression in lung,in thymus,spleen,intestine with OVA sensitization model.3.Though skin sensitization might be an important way in the allergic disease in infants.
