Pretreatment Of IL-2 Influences The Proliferation And Differentiation Of Mice Splenic Lymphocyte Following Mobilization With G-CSF | Posted on:2010-12-03 | Degree:Master | Type:Thesis | Country:China | Candidate:H S He | Full Text:PDF | GTID:2144360275991321 | Subject:Internal Medicine | Abstract/Summary: | PDF Full Text Request | Objectives:To explore the influence of IL-2 pretreatment on splenic lymphocyte following mobilization with G-CSF,which may provide a new approach to attenuate aGVHD.Methods:1.Splenic cells and naive CD4+T cells from C57BL/6 mice receiving G-CSF-mobilized were coclutured with the allogeneic antigens from BALB/c mice.The proliferation responses and the differentiation of T cells were determined.2.Splenic cells from C57BL/6 mice were pretreated with IL-2 and the expression of SOCS-3 gene was detected.3.Splenic cells and naive CD4+T cells from C57BL/6 mice were pretreated with 50U/mL IL-2,and coclutured with the allogeneic antigens from BALB/c mice.The proliferation responses and the differentiation of T cells were determined.4.Splenic cells and naive CD4+T cells from C57BL/6 mice receiving G-CSF-mobilized were pretreated with IL-2(50U/mL),and coclutured with the allogeneic antigens from BALB/c mice.The proliferation responses and the differentiation of T cells were determined.Results1.The proliferation responses of splenic cells and naive CD4+T cells from C57BL/6 mice receiving G-CSF-mobilized to allogeneic antigens were significantly abrogated.the differentiation of T cells were polarized toward the production of type-2 cytokines.2.The expression of SOCS-3 in C57BL/6 mice splenic cells were dramatically elevated by the pretreatment with IL-2,which reached the peak after 6h pretreatment3.The proliferation responses of splenic cells and naive CD4+T cells from C57BL/6 mice to the allogeneic antigens were significantly abrogated by the pretreatment with IL-2,the differentiation of T cells were polarized toward the production of type-2 cytokines.4.The proliferation responses of splenic cells and naive CD4+T cells from C57BL/6 mice receiving G-CSF-mobilized to the allogeneic antigens were significantly abrogated by the pretreatment with IL-2,the differentiation of T cells were polarized toward the production of type-2 cytokines.The combination of G-CSF and IL-2 is potentially synergetic in the induction of T lymphocyte immune tolerance.
