PartⅠStudy on differences of neutral oligosaccharides in breast milk between Chinese and American mothersObjectiveTo investigate the concentration of oligosaccharides in Chinese mothers' breast milk and its pattern of change at different time points;analyze the proportion of different oligosaccharides to total oligosaccharides concentration;to find out the differences of oligosaccharides at three time points through comparison with milk of American mothers'.Methods51 pairs of Chinese infant and mother were recruited in this study.Breast milk was collected at 2,4,13 week postpartum and saliva sample at 2 week.American mothers were recruited according to the same standards.Neutral oligosaccharides concentration of breast milk was analyzed by High-performance liquid chromatography. Results1,There were not significant changes in the total concentration of neutral oligosaccharides in Chinese maternal breast milk postnatal 13 weeks,but the concentration of different types of neutral oligosaccharide varies at each visiting interval.The concentration of 3-FL and 2'-FL increased from week 2 to week 4 (84.6-485.8μg/ml;579.6-787.8μg/ml;P<0.05),but LNF-1 decreased from 596.9mg/ml to 304.5mg/ml2,According to the classification of the neutral oligosaccharides,29 mothers are Le(a-,b+),and 10 of them are Le(a-,b-) and 12 of them are Le(a+,b-)3,There were significant changes in the total concentration of neutral oligosaccharides in American maternal breast milk postnatal 13 weeks.LNF-1 decreased in the first 13 weeks(P<0.05).The concentration of 3-FL increased(P<0.05). The concentration of 2'-FL,LDFH-Ⅰ,LNF-2,LNF-4 and LDFH-Ⅱwas stable.4,There were significant differences between Chinese and American maternal breast milk in total oligosaccharides concentration at 2,4,13 weeks.5,There was significant difference in concentration ofα1,2 linked oligosaccharides between Chinese and American maternal breast milk at 2 weeks.But there were no significant differences at 4 and 13 weeks.6,The concentration ofα1,3 linked oligosaccharides in American maternal breast milk was significantly higher than that in Chinese maternal breast milk at 2,4 and 13 weeks.Conclusion:1,The variation of neutral oligosaccharide in Chinese mothers' breast milk is unique.The expression ofα1,2 linked oligosaccharides decreased;the expression of α1,2 linked oligosaccharides increased gradually in the first 13 weeks postpartum.2,The distribution of Lewis/Sec genotype matches with the distribution of general people group in China3,The concentration of neutral oligosaccharides in American maternal breast milk was significantly higher than that in Chinese maternal breast milk;the concentration ofα1,3 or 1,4 linked oligosaccharides was much higher in American maternal breast milk than that in Chinese maternal breast milk;there were no significant difference in the concentration ofα1,2 linked oligosaccharidesPartⅡGene polymorphism of FUT2 and its relationship with oligosaccharides concentration in American and Chinese mothersObjectiveIn order to explore the relationship between oligosaccharides differences and gene polymorphism,SNP spot on gene FUT2 was assayed and oligosaccharides concentration was analyzed based on SNP genotype.MethodSaliva were collected using Oragene KIT.DNA was abstracted from saliva and used for SNP assay.Result1,It can be A or G on SNP(rs601338).There are three genotypes as AA,AG, and GG.12 of American mothers are AA;24 of them are AG;15 of them are GG. 2,There are moreα1,2 linked oligosaccharides in genotype GG maternal breast milk than in genotype AA;there are moreα1,3 linked oligosaccharides in genotype AA maternal breast milk;the concentration ofα1,2 linked oligosaccharides andα1,3 linked oligosaccharides in genotype AG maternal breast milk lay between AA and GG.3,All of the 50 Chinese mothers are GG type.There were no obvious relationship between SNP genotype and neutral oligosaccharides concentration in this group.Conclusion1,There exists gene polymorphism of FUT2 SNP2,There was a clear relationship between SNP genotype and oligosaccharide concentration in American group.3,There was no definite relationship between SNP genotype and oligosaccharide concentration in Chinese group.
