ObjectiveIn this experiment,in order to study the effect of the pore size on the growth of Schwann cell in silk fibroin scaffold,the growth and proliferation of Schwann cell in silk fibroin scaffold with different pore size was characterized by SEM observation.Material and Method:1.Prepare silk fibroin scaffolds with different pore size,i.e.larger pore size scaffolds(pore size 50~60μm),smaller pore size scaffolds(pore size 10 20μm),each of three samples.2.Schwann cells(R3[33-10ras3]) as the seed cells were incubated in 37℃,5%CO2 incubation box.When cells filled the bottom to form a dense monolayer cells in cell culture bottle,we digested the cells,used 5%FBS to adjust the cell concentration to 1x106/ml,and then seeded Schwann cells in the porous silk fibroin scaffolds with different pore size in vitro.3.After one week of co-culture the cell morphological observation was characterized by SEM.Results:On the material with smaller pore size,the phenotype of cell was bipolar, and the cells adhered onto the surface.On the material whose pore size was significantly larger than the cell size,most cells were spherical,some of which were single scattered on the surface of porous scaffolds,while others became into cell cluster at the bottom.But some of the cells were still bipolar on the the ridge between the pores.The cell density on scaffolds with larger pore size is higher than those with smaller pores. Conclusion:The results showed that the topography of silk fibroin scaffold is an influential factor for growth of Schwann cell.Schwann cells prefer to grow on the ridge between the pores.According to this case,we can design more suitable scaffold for growth of Schwann cells in future.
