Effect Of Cholesterol-bile Acid Metabolism In New Zealand White Rabbit Fed By Cholesterol-rich Diet With Fufang Jiangzhi No 3 | | Posted on:2010-08-29 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q Q Mao | Full Text:PDF | | GTID:2144360275991822 | Subject:Internal Medicine | | Abstract/Summary: | PDF Full Text Request | | Part 1 Effect of Fufang Jiangzhi No 3 on the CHO,LDL-C and BA change in New Zealand White Rabbit fed by cholesterol-rich dietObjective To evaluate the Effect of Fufang Jiangzhi No 3 on the CHO,LDL-C and BA change in New Zealand White Rabbit fed by cholesterol-rich diet.Methods 24 male healthy New Zealand white rabbit were randomly assigned into three groups from group A to group C:control group(group A),hypercholesterolemia model group(group B),Fufang Jiangzhi No 3 intervention group(group C).Group B and group C were fed by cholesterol-rich diet(contain 1%cholesterol) 120g/day during 4 weeks' observation in order to induce hypercholesterolemia while the group A was fed by common rabbit fodder 120g/day.Group C recieved Fufang Jiangzhi No 3 intragastic administration(0.5bag/20ml distilled water,every morning) at the same time as the start of the cholesterol-rich diet exposure.Serum CTKK LDL-C and BA assessment of the rabbits fed by cholesterol-rich diet was performed at the end of the experiment.Results The Serum CHO in group B and group C showed a increasing tendency. The Serum CHO in group B and group C were much higher than those in the group A(P<0.01),moreover the Serum CHO in group C was lower than those in group B(P<0.01).The Serum LDL-C in group B and group C showed a increasing tendency. The Serum LDL-C in group B and group C were much higher than those in the group A(P<0.01),moreover the Serum LDL-C in group C was lower than those in group B(P<0.01).The Serum BA in group B and group C showed a increasing tendency. The Serum BA in group B and group C were much higher than those in the group A(P<0.01).The difference between group B and group C has no significance(P>0.05).Conclusions Fufang Jiangzhi No 3 intervention can attenuate serum CHO and LDL-C of cholesterol-rich diet-induced hypercholesterolemia in New Zealand white rabbit.Part 2 The effect of Fufang Jiangzhi No 3 on the expression of CYP7A1 mRNA and the activity of CYP7A1 in hepatic tissue in New Zealand White Rabbit fed by the cholesterol-rich dietObjective To evaluate the the effect of Fufang Jiangzhi No 3 on the expression of CYP7A1 mRNA and the activity of CYP7A1 in hepatic simple in New Zealand White Rabbit fed by cholesterol-rich diet and approach the mechanisms of the preventive effect on cholesterol-bile acid metabolism of Fufang Jiangzhi No 3.Methods Animals were grouped and intervened as described above.The expression of CYP7A1 mRNA was measured by realtime PCR.The activity of CYP7A1 was measured by ELISA.Results The expressions of CYP7A1 mRNA in hepatic simples of three groups were,respectly,1.97±1.50,0.70±0.27 and 3.87±1.68.The level of group B was much lower than that of group A(P<0.01).The level of group C was higher than that of group B and group A(P<0.01).The concentrations of CYP7A1 in hepatic simples of three groups were,respctly,0.80±0.22μg/ml,0.44±0.16μg/ml and 0.81±0.21μg/ml.The concentration of group B was much lower than that of group A(P<0.01).The concentration of group C was much higher than that of group B(P<0.01). The difference between group A and group C has no significance(P>0.05).Conclusions The results suggested:the cholesterol-rich diet can inhibit the expression of CYP7A1 mRNA in the liver.Fufang Jiangzhi No 3 can attenuate the inhibition of cholesterol-rich diet,thereby,it can promote the convertion of cholesterol to bile acid.The level of cholesterol decreased.Part 3 The effect of Fufang Jiangzhi No 3 on the expressions of BSEP mRNA,SHP mRNA and LDL mRNA in hepatic tissue in New Zealand White Rabbit fed by cholesterol-rich dietObjective To evaluate the the effect of Fufang Jiangzhi No 3 on the expression of BSEP mRNA,SHP mRNA and LDL mRNA in hepatic tissue in New Zealand White Rabbit fed by cholesterol-rich diet and explore the mechanisms of the preventive effect of Fufang Jiangzhi No 3.Methods Animals were grouped and intervened as described in Part 1.The expressions of BSEP mRNA,SHP mRNA and LDL mRNA were measured as described in Part 2.Results The expressions of BSEP mRNA in hepatic tissue of three groups were,respectly,7.33±2.44,16.40±3.01 and 9.64±1.63.The expressions of SHP mRNA in hepatic tissue of three groups were,respectly,9.83±2.50,85.44±6.79 and 20.73±4.47.The level of BSEP mRNA and SHP mRNA in group B was much higher than those of group A(P<0.01).The level of group C was lower than those of group B(P<0.01).The level of group C was higher than those of group A(P<0.01).The expressions of LDL mRNA in hepatic tissue of three groups were,respectly,5.76±1.96,3.29±1.58 and 9.38±2.86.The level of group B was much lower than that of group A(P<0.01).The level of group C was higher than that of group B(P<0.01).The level of group C was higher than that of group A(P<0.01)Conclusions Fufang Jiangzhi No 3 can attenuate the activation of FXR,downregulate the expressions of BSEP mRNA and SHP mRNA.It may be one of the mechanisms involved in its preventive effect in cholesterol-rich diet-induced hyperchlesterolemia in New Zealand White Rabbit. | | Keywords/Search Tags: | cholesterol, bile acid, hypercholesterolemia, cholesterol-rich diet, FXR, Fufang Jiangzhi No 3, BSEP mRNA, SHP mRNA, LDL mRNA | PDF Full Text Request | Related items |
