ObjectiveThis topic is based on the previous study of clinical and animal experiments. Through the establishment of PSD rat model and acupuncture Baihui,Fengfu,Taichong,Shenmen, by using Real-time Quantitative polymerase chain reaction Quantitative(Real-time PCR)to observe the Norepinephrine Receptor2R(NEa2R), 5-hydroxytryptamine Receptor1A(5-HT1AR) and 5-hdroxtryptamine transporter(5-HTT) expression of m RNA in the PSD rat brain hippocampus, raphe nucleus, blue spot. From molecular level, to further explore the mechanism of acupuncture intervention on post-stroke depression. To promote the clinical application of acupuncture in the treatment of PSD, enrich the theory of acupuncture, has a very important practical significance and scientific significance. Method:120 SD rats of SPF grade were selected, the month old was 3 ~ 5 months old,the male body weight was 220~250g, giving the adaptive feeding of the standard mixed feed.The experiment was carried out after 1 weeks. The rats were randomly divided into blank group,model group,drug group and acupuncture group.Among them,the blank group is 10, model group is 30, drug group and acupuncture group is 40. PSD rat model was made by cerebral artery occlusion MCAO(middle cerebral artery occlusion) with CUMS(chronic unpredictable mild stress).Blank group were given normal diet;model group after modeling succeed but did not give any treatment;drug group were given fluoxetine hydrochloride 2mg/kg dissolved in saline gavage,1 time a day,7 days for a course of treatment, the total was three courses.In the middle of each course rest 1 day;acupuncture group selected Baihui,Fengfu,Shenmen,Taichong.Used Hua Tuo brand 0.20x13 mm stainless steel needle.The needled depth is about 2mm,mild twirling needle retention during 20 min,1 time a day,7 days for a course of treatment, the total treatment was 3 courses, during each course rest 1 day. After the completion of the experiment, we used the method of Quantitative Real-time PCR to detect the content of Norepinephrine Receptor2R(NEa2R),5-hydroxytryptamine Receptor1A(5-HT1AR) and 5-hdroxtryptamine transporter(5-HTT)in the PSD rat brains hippocampus, raphe nuclei and locus coeruleus.The analysis method used in this experiment: relative quantification study, calculated as 2- Delta Ct. The experimental measured data was used the mean ± standard deviation(x± s), Within the groups mean compared using paired t test, different groups mean compared using analysis of variance(ANOVA).Statistical results were analyzed by computer software(Spss for windows17.0).There is the significant differences when p < 0.05, there is the very significant differences when p< 0.01.Statistical chart drawing by EXCEL 2000 software. Result: 1 The expression of Hippocampal Monoamine Neurotransmitter and receptor m RNA:Changes of 5-HTT m RNA expression:Compared with the blank group, the expression of model group about 5-HTT m RNA in the hippocampus was significantly reduced(p<0.01); Compared with the model group, acupuncture group's and drug group's the expression of the 5-HTT m RNA in hippocampus was significantly increased(p<0.01). The difference was statistically significant. Compared with the drug group, the acupuncture group's expression of 5-HTT m RNA in hippocampus was significantly increased(p>0.05),the difference was not statistically significant.Changes of 5-HT1 A receptor m RNA expression:Compared with the blank group, the expression of model group about expression of 5-HT1 A receptor m RNA in the hippocampal was significantly reduced(p<0.01); Compared with the model group, acupuncture group's and drug group's expression of 5-HT1 A receptor m RNA in the hippocampus was significantly increased(p<0.01), the difference was statistically significant. Compared with the drug group, acupuncture group's expression of 5-HT1 AR m RNA in the hippocampus increased was significantly(p>0.05), the difference was not statistically significant.NEa2R m RNA expression changes of: Compared with blank group, the expression of model group about NEa2 R m RNA in the hippocampus was significantly increased(p<0.01); compared with the model group, acupuncture group's and drug group's expression of NEa2 R m RNA in hippocampus was significantly reduced(p<0.01). Compared with the drug group, acupuncture group's expression of NEa2 R m RNA in the hippocampus increased significantly(p> 0.05), the difference was not statistically significant. 2 The expression of raphe nucleus monoamine neurotransmitter and receptor m RNA:Changes of 5-HTT m RNA expression: Compared with the blank group, the expression of model group about 5-HTT m RNA in the raphe nuclei was significantly reduced(p<0.01); Compared with the model group, acupuncture group's and drug group's the expression of the 5-HTT m RNA in raphe nuclei was significantly increased(p <0.01). The difference was statistically significant. Compared with the drug group, the acupuncture group's expression of 5-HTT m RNA in raphe nuclei was significantly increased(p<0.05),the difference was statistically significant.Changes of 5-HT1 A receptor m RNA expression: Compared with the blank group, the expression of model group about expression of 5-HT1 A receptor m RNA in the the raphe nuclei was significantly reduced(p<0.01); Compared with the model group, acupuncture group's and drug group's expression of 5-HT1 A receptor m RNA in the raphe nuclei was significantly increased(p<0.01), the difference was statistically significant. The expression of locus coeruleus monoamine neurotransmitter and receptor m RNA:Changes of 5-HTT m RNA expression: Compared with the blank group, the expression of model group about 5-HTT m RNA in the locus coeruleus was significantly reduced(p<0.01); Compared with the model group, acupuncture group's and drug group's the expression of the 5-HTT m RNA in the locus coeruleus was significantly increased(p<0.01). The difference was statistically significant. Compared with the drug group, the acupuncture group's expression of 5-HTT m RNA in the locus coeruleus was significantly increased(p<0.05),the difference was statistically significant.Changes of 5-HT1 A receptor m RNA expression:Compared with the blank group, the expression of model group about expression of 5-HT1 A receptor m RNA in the locus coeruleus was significantly reduced(p<0.01); Compared with the model group, acupuncture group's and drug group's expression of 5-HT1 A receptor m RNA in the locus coeruleus was significantly increased(p<0.01), the difference was statistically significant. Compared with the drug group, acupuncture group's expression of 5-HT1 AR m RNA in the locus coeruleus increased was significantly(p>0.05), the difference was not statistically significant.NEa2R m RNA expression changes: Compared with blank group, the expression of model group about NEa2 R m RNA in the locus coeruleus was significantly increased(p<0.01); compared with the model group, acupuncture group's and drug group's expression of NEa2 R m RNA in the locus coeruleus was significantly reduced(p<0.01). The difference is statistically significant. Compared with the drug group, acupuncture group's expression of NEa2 R m RNA in the locus coeruleus increased significantly(p>0.05), the difference was not statistically significant.Conclusion:1 The expression of NEa2 R m RNA in the brain of model group increased, the expression of 5-HTT and 5-HT1 AR decreased. It suggested that the transport rate of NEa2 R, 5-HTT, 5-HT1 AR with 5-HT, NE in the brain of PSD rats could be changed..2 Acupuncture can reduce the expression of monoamine neurotransmitter NEa2 R in the brain of PSD rats, and increase the content of 5-HTT and 5-HT1 AR.Revealed that the mechanism of acupuncture about treatment of PSD maybe related to improve the rat brain monoamine neurotransmitter transport rate of 5-hydroxytryptamine(5-HT) and norepinephrine(NE).3 The general expression of NEa2 R m RNA, 5-HTT m RNA and 5-HT1 AR m RNA in rat brain between acupuncture group and drug group have a little difference. Acupuncture group raphe nucleus, locus coeruleus of 5-HTT m RNA expression were increased more than the drug group, suggesting that the acupuncture in the treatment of PSD has some certain advantages.These results indicate that significant effect of acupuncture treatment of PSD rats, we hypothesized that the possible mechanism: acupuncture can significantly improve the model rat brain monoamine neurotransmitter 5-hydroxytryptamine(5-HT) and Norepinephrine(NE) content or utilization. This provides a reliable experimental basis for clinical application of acupuncture therapy in the treatment of PSD.
