ObjectiveThe aim of this study was to analyse the serum PSA concentratione insulin,testosterone,the body mass index of the adolescents with polysystic ovary syndrome,further to understand these markers in the adolescents with polycystic ovary syndrome,Is prostate specific antigen a potential new marker of androgen excess in polycystic ovary syndrome;To study X chromosome inactivation pattern of PCOS,whether X chromosome inactivation pattern influences susceptibility to PCOS,with human androgen receptor gene(HUMARA)analysis,to provide evidence in order to correctly diagnose and therapy the PCOS in adolescents.Methods28 case with PCOS in adolescents diagnosed in epidemiological survey about adolescents in Tianjin were selected,between March in 2007 and March in 2008,according the ESHRE/ASRE criteria.1.Body height and body weight were measured,the body mass index(BMI) were caculated according to the formular(body mass index=body weight(kg)/(body height)~2(m~2)).serum PSA,insulin,glucose,testosterone were measured using biochemical equipment,then these parameters of PCOS were compared to the control groups.2.X chromosome inactivation analysis(1) DNA was extracted using whole blood from the patients with PCOS and the controls,and stored at -20℃.(2) Restriction enzyme treatment.(3) Nested PCR amplification.(4) PCR products were run on gel.Results(1) Five of the 28 PCOS(17.9%) had a regular menstrual cycle and 23(83.1%) had a irregular menstrual cycle,including of oligomenorrhea(57.1%),secondary amenorrhea(21.4%) and dysfunctional uterine bleeding(3.6%).The obese group had the longer menstrual cycle than the non-obese group,it shows significant difference between the two groups(P<0.05).(2) The mean BMI,serum testosterone(T),prostate specific antigen(PSA) and Homeostasis Model Assessment(HOMA) levels were found to be significantly higher in patients with PCOS than healthy controls in adolescents(P<0.05).(3) The rate of obesity,insulin resistance,hyperandrogenism of PCOS in adolescents respectively was 28.6%,21.4%,85.7%,and the mean BMI was higher in IR group than in non-IR group(P<0.05).(4) Serum PSA levels were positively correlated with FG scores(r=0.417,P<0.05),serum testosterone(r=0.551,P<0.05).(5) The rate of X chromosome norandom inactivation in PCOS and control groups were 8.33%and 4%,there was no significant difference between the PCOS group and the control group(P>0.05).Conclusion(1) The mean levels of serum PSA were significangtly higher in adolescents with PCOS when compared to the healthy control groups.Serum PSA levels were positively correlated with FG scores and serum testosterone,and it may be a potential new marker for androgen excess in patients diagnosed as PCOS(2) X chromosome inactivation patterns were found to be no significant difference between the PCOS and the control groups in adolescents.
