| Objective: To investigate the effects of recomdination human growth hormone(rhGH) on proliferation of alveoliar typeâ…¡cells and expression of Bcl-2 and Bax in rats with smoke inhalation injury.Methods: Seventy Sprague-Dawley rats weighting 180-220g were randomly divided into three groups:group I was normal controles; groupâ…¡was smoke inhalation combined with 0.9% saline administraion (30);groupâ…¢was smoke inhalation combined with rhGH administraion(30). The animals in groupâ…¡andâ…¢were sacrificed at 6,10,14 day after smoke inhalation injury. RhGH(1.33u/kg) was administered subcutaneously in rhGH group for 7days. The rats were killed and the lungs were removed separately at 6d,10d and 14d,observed the proliferation of ATâ…¡cells and expression of Bcl-2 and Bax protein by immunohistochemical method.Results: The rats that had suffered from inhalation injury appeared dysphoric, had tachypnea and tachycardia. The rats in control group appeared to have stable respiration. Groupâ…¡was found to have severe diffused and passive lung congestion. There was no obvious sign of edema or spot bleeding in lungs in groupâ…¢. Morphological studies found that there were more proliferated ATâ…¡cells in groupâ…¢than groupâ…¡.The rats in normal control group have no expression of Bcl-2 and Bax protein. However, there was obvious expression of Bcl-2 and Bax in rats with smoke inhalation injury. In groupâ…¡,the expression of Bcl-2 and Bax was all increased after inhalation injury. The expression of Bcl-2 increased but Bax protein decreased from the beginning of 6d treated with rhGH in the treatment groupâ…¢,the differences have obvious statistical significance.Conclusions: RhGH can stimulate the proliferation of ATâ…¡cells, and up-regulate the expression of Bcl-2,and down-regulate the expression of Bax and change the ratio of Bcl-2/Bax to decrease apoptosis of the lung cell. |
