Objective:With living standards improved,the consumption of alcohol and the number of alcoholics increases year by year.Chronic alcoholism have become a serious impact on human health.Alcohol has the potential biological effects on almost every organ system of people.The impact of alcohol on the lung has a great clinical importance,for its volatility makes airway epithelium in high level ethanol surroundings.It can cause airway epithelial injuried,evoke and aggravate alcohol-related pulmonary diseases.At present,the research about alcohol-induced pulmonary fibrosis is few.Therefore,the study aims to make the model of pulmonary fibrosis in rats,and observe the effects of alcohol intake on normal lung and pulmonary fibrosis.To explore the relationship between alcohol and pulmonary fibrosis and investigate the possible mechanism for the pathogenesis of pulmonary fibrosis by observing transforming growth factor-β1,glutathione,hydroxyproline expression.It is significant to find the relationship between alcohol and pulmonary fibrosis,and to intensify the propaganda of the pemiciousness of alcohol taking.Methods:Clean healthy male Sprague-Dawley rats 32,weighing 180-220g,were randomly divided into control group(C group)8,tracheal saline injection;alcohol group(A group) 8,tracheal saline injection and alcohol gavage 4-Sg/kg/day, bleomycin group(B group) 8,5mg/kg tracheal bleomycin injection,alcohol+ bleomycin group(AB group) 8,alcohol and oral tracheal bleomycin 5mg/kg injection. Corresponding treatment of rats in each group after the diet are water freely.Animals of each group were killed 6 weeks after the anesthesia,thoracotomy complete separation of the trachea and lung tissue.Then the lung weight was recorded and the lung index was re-calculated.By hematoxylin-eosin staining the morphological changes were observed lung tissue,by Masson staining interstitial lung collagen deposition were observed.Lung tissues of transforming growth factor-β1 expression in rats were detected through Elisa;glutathione and hydroxyproline in lung tissue were measured through colorimetric.SPSS16.0 statistical software were used for data processing,parameter data to mean±standard deviation,using of t test and one-way ANOVA..The level of significance was set as P<0.05. Results:1.The general status:Before the experiment,the general condition of rats was normal,and glossy fur was smooth.After the experiment,Each group was no significant differences in body weight,but irrigation alcohol rats growed slowly, gloomy,moved slowly and have soft row of faceless stool.2.HE staining showed that: the control group alveolar structure was clear,and alveolar walls interstitial hadn't inflammatory cell infiltration.Alcohol group and bleomycin group can be seen varying degrees of alveolar and alveolar septal infiltration of inflammatory cells, some alveolar wall damaged or collapsed.Masson results show:the experimental group had more deposition of collagen fibers at intervals of alveolar than the control group,especially alcohol-bleomycin group.3.Transforming growth factor-β1 in the C group than A group,B group,AB group are statistically significant,all P<0.05.It showed that chronic alcohol consumption and intratracheal injection of bleomycin can make the lung tissue TGF-β1 expression increase.TGF-β1 had statistical significance between the AB group and B group,P<0.05.Chronic drinking can make the lung tissue TGF-β1 expression increase.Glutathione(GSH) and hydroxyproline(HYP) in the C group and A group,B group,AB group were statistically significant(P<0.05).Chronic alohol intake,intratracheal injection of bleomycin in the lung tissue both can make the expression of GSH reduces,HYP increases.Conclusion:This experiment proved intratracheal injection ofbleomycin induced lung injury and caused the formation of pulmonary fbrosis,so it can be served as a study of a model of pulmonary fibrosis.Alcohol can cause the reduction of glutathione in lung tissue,make the defense system of lung tissue damage;on the basis of alcohol induced to produce TGF-β1 and the lung tissue content of hydroxyproline increased,it was proved that alcohol play an important role on the formation of pulmonary fibrosis,suggesting that chronic drinking can cause lung injury in rats,leading to pulmonary fibrosis.Alcohol may be one of the causes of pulmonary fibrosis.
