Objective:To analyse the loss of heterozygosity(LOH) and microsatellite instability(MSI) at partial sites of chromosome 3p and 4p in human cervical carcinoma, and investigate their roles in the development of human cervical carcinoma.Methods:LOH and MSI with 6 microsatellite sites and infection of human papilloma virus(HPV)16 in 59 invasive carcinoma of cervix cases and 49 cervical intraepithelial neoplasia(CIN) cases were analyzed by using PCR-denaturing polyacrylamide gel-silver staining method.Partial sites were detected by gene sequencing.Results:1.There were significant difference in LOH rate among lower-level CIN(CIN1), higher-level CIN(CIN2,3) and invasive carcinoma of cervix at D3S1234,D3S1300 and D3S1289(P<0.05 ).There were significant difference in LOH rate among their respective histological degree at D3S1234,D3S1300 and D3S1766(P<0.05).2.No significant difference were observed in MSI rate between invasive carcinoma of cervix and CIN at all the sites(P>0.05).3.There was no significant difference between the infection rate of HPV16 and LOH rate in invasive carcinoma of cervix or CIN(P>0.05).4.The gene sequencing to D4S412 revealed that its base transformation happen at a constant position.Conclusion:1.The LOH of 3p and the FHIT gene in which is an early event in cervical carcinoma;The 4p LOH happened at early and late stage.Therefore,the detecting of LOH rate at multisites play an important role in the early diagnosis,treatment and the estimation of prognosis in cervical carcinoma. 2.MSI in cervical carcinoma is low-frequency case,which has no significant role in prediction in the early diagnosis of cervical carcinoma.3.The HPV infection in the progress of cervical lesions is important,but only HPV infection is not sufficient to lead to malignant transformation.4.The gene sequencing in this issue should not come to more definite conclusions, because of the small sample size,smaller sites,and also not yet seeing the relevant report, but gene sequencing still has broad prospects.
