Background Psoriasis is immunologic abnormality and polygenic inheritance skin disease, is a chronic inflammatory dermatosis. The key histopathology event is keratinocyte proliferation, inflammatory cell infiltrate in dermis, expansion and dilation of superficial dermal microvasculature .The incidence rate is disparation in different area. It affects approximately 0.1~6.5% of the population all the world and 0.123% in China. Psoriasis vulgaris take percent more than 90% of all psorisis patients. It contain drop psoriasis and plaque psoriasis (coin type, map type and crust form).The mechanism of psoriasis has not been entirely cleared, it's general consider that psoriasis is a complicit disease cause by inheritance and environmental factor.Recent studies have shown the Single nucleotide polymorphism of VEGF which located on chromosome 6p21 has association with psoriasis invasion. VEGF could increase permeability of capillary vessel, promote endothelial cell Division, generation, promote neovascularization. It takes an important role in vascularization. VEGF not only promote dermis vascular endothelial cell Division and generation, but also increase the permeability of capillary vessel, and make a chemistry chemotaxis role to inflammatory cell and endothelial cell. Aggravate the inflammatory reaction in psoriasis lesion, promote the patho-process of psoriasis. Although most researches on VEGF association with psoriasis invasion were carried out on the world, genotype/phenotype correlations are still unknown.Objective To investigate the association between single nucleotide polymorphism (SNP) (rs2010963, rs9367178) in vascular endothelial growth factor (VEGF) gene and psoriasis susceptibility in Chinese Han population, and investigate the possible Pathogenesis of psoriasis.Methods Based on the results of genome wide association study of psoriasis which were performed by our team, we used the Sequenom MassArray system to genotype two single nucleotide polymorphism (rs2010963, rs9363178) of VEGF gene in 1,103 psoriasis cases and 1,104 normal controls, and then we used the Plink 1.03 software for statistical analysis.Results There were no significant differences in allele frequencies at the SNP rs2010963(P=0.155) or rs9367178(P=0.65)sites when comparing all psoriasis patients (1,103) with controls(1,104). And, when patients were distribute in subgroup. There were no significant differences in allele frequencies at the SNP rs2010963 or rs9367178 sites when comparing each group with controls too(P>0.05).Conclusion It suggested that SNPs rs2010963 and rs9367178 of VEGF gene could not related with psoriasis susceptibility in Chinese Han population.
