Objective:The model of rats with severe acute pancreatitis-associated lung injury by this study,The use of TNF-αinhibitor for their intervention, comparing the sham-operated group, SAP group and intervention group in all three group.In order to evaluating the effect of on severe acute pancreatitis,pancreatic cells apoptosis ,Caspase-3 expression and lung injury in rats by TNF-αinhibitor.Methods:Detection of serum amylase:The detecting of serum amylase by CX9 automatic biochemical analyzer;Detection of TNF-α: The use of ELISA method;RT-PCR experiment: detection of ET-1mRNA and ICAM-1mRNA in lung tissues;Caspase-3, ET-1 and NF-κB protein expression tests: ET-1,NF-κB expression in lung tissue and Caspase-3 expression in pancreatic tissue by two-step method using S-P staining;Detection of apoptotic cells: Pancreatic tissue and lung tissue apoptosis by TUNEL, calculated 100 pancreatic cells in the percentage of positive cells, namely, apoptosis index;lung tissue wet/dry ratio: thoracotomy immediately removed the right lobe of the lung using an electronic balance, said after the wet weight at constant temperature oven to constant weight to calculate the wet/dry ratio;Histopathological observation of the pancreas: pancreatic tissue score principle to its score according to Gross by HE staining;Results:The successful construction of the rats with severe acute pancreatitis-associated lung injury model; For serum amylase and TNF-α, sham-operated group was significantly lower than SAP group and intervention group; SAP group and intervention group were higher than those of the intervention group;Pancreas in sham-operated group, Caspase-3 protein has no obvious expression of the pancreas. In the intervention group, Caspase-3 protein expression was higher than the SAP group; sham operation group of the pancreatic cells and lung tissue cells, there is no obvious apoptotic cells; pancreatic cells and lung tissue cell apoptosis in SAP group is lower than the intervention group;The expression of ET-1,NF-κB and ICAM-1 was not obvious in lung tissue of sham-operated group, the expression level of ET-1 and NF-κB in the SAP group was significantly higher than the intervention group;The lung tissue wet / dry ratio of sham group was significantly lower than the intervention group and SAP group, lung tissue wet / dry ratio in the SAP group was higher than the intervention group;sham-operated group of pancreatic tissue necrosis was not obvious, the extent of pancreatic necrosis in the SAP group is higher than the intervention group.Conclusion:TNF-αinhibitor can significantly reduce the inflammatory response of the pancreatic tissue and lung tissue in rat with severe acute pancreatitis, reduce serum amylase and TNF-α, increased the expression of Caspase-3 protein pancreatic tissue, reduce the expression of NF -κB,ICAM-1 and ET-1,reduce wet-dry ratio of the lung tissue, increased cell apoptosis of lung tissue and pancreatitis tissue and improve lung microcirculation to reduce inflammation injury to the pancreatic tissue.
