Objective To investigate the expression of Endocan in the marrow of acute leukemia and its clinical significance and the effects of 4-HPR on the acute leukemia cell line NB4 in vitro and the potential mechanisms. Methods The Endocan expression in the marrow of 33 Acute leukemia patients was detected by Immunocytochemical assay(S-P method) and Western blot. MTT method was used to assess the antiproliferation effect of 4-HPR. Flow cytometry was used to study the effects on cell cycle and apoptosis.Western blot was used to detect Endocan and retinoid acid receptor expression after treated with 4-HPR. Results The Endocan was expressed in 20 of 33 patients with acute leukemia. The expression rate is 60.6%. And the Endocan and Opn expression was significantly down regulated when released after chemotherapy compared with before chemotherapy. 4-HPR inhibited NB4 proliferation effectively with a concentration-dependent manner and decreased the expression of Endocan, Opn and VEGF.4-HPR could obviously the expression of retinoid acid receptor RARαand RXRγ. It alsounregulate RXRβexpression but has no effect on the expression of retinoid acid receptor RARβ,RARγ和RXRα. Conclusion Endocan may play an important role in the genesis and development of acute leukemia and it might be a potential feature of prognosis of acute leukemia. 4-HPR could inhibit the proliferation of acute leukemia cell remarkably and could downregulate Endocan, Opn and VEGF expression. 4-HPR may play its roles by retinoid acid receptor RARαand RXRγassociated pathway.
