Objective: To investigate the effects of the acetoacetate extract of Vitex negundo seed on the growth of human cervical cancer Hela cells and its mechanism in human cervical cancer Hela xenograft nude mouse model.Methods: Human cervical cancer Hela cell line was cultured in vitro. Proliferation of Hela cells treated with 6 species of the extracts from Vitex negundo seed using various methods of extraction and fractionation was measured by MTT assay. The established tumor xenograft in nude mouse model was used to evaluate therapeutic effect of the extract of Vitex negundo seed with the most inhibiting proliferation efficacy in human cervical cancer Hela cells. SP immunohistochemical method was used to analysis expression of proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor(VEGF) in nude mouse human cervical cancer Hela xenografts.Results: Six species of the extracts from Vitex negundo seed inhibited proliferation of Hela cells, in a concentration-dependent manner. The acetoacetate extract of Vitex negundo seed (EVn-50) is the strongest of those, Its IC50 value is 5.5μg/mL. The inhibition rates of tumor growth and tumor weight were 26.7%, 31.8%, 52.5%and 40.0%, 49.6%, 54.5% respectively after treated with EVn-50 (5mg/kg,10mg/kg,20mg/kg). Expression of PCNA and VEGF protein in human cervical carcinoma xenograft cells were down-regulated.Conclusion:1. EVn-50 significantly inhibits the proliferation of the human cervical cancer Hela cells, in a concentration-dependent manner. 2. EVn-50 can suppress the growth of human cervical cancer Hela xenografts in nude mouse model, in a dose-dependent manner.3. Therapeutic action of EVn-50 on human cervical cancer Hela xenografts in nude mouse model may be associated with the down-regulation of PCNA and VEGF protein.
