Objective: To investigate the roles of lovastatin on the proliferation and apoptosis of the human colon cell line HCT-116, also to explore its possible mechanisms of anticancer effects.Methods: In vitro, human colon cancer cell line HCT-116 was treated by lovastatin in various concentrations(5μmol/l,10μmol/l,20μmol/l,40μmol/l). By means of MTT assay, the absorbance value was detected and growth inhibition rate was calculated .Cell cycle and apoptosis was assessed by flow cytometry. the expression of bcl-2 and bax and MMP9 were analyzed by Western Blot.The expression of MMP9mRNA was determined by reverse transcriplase polymerase chain reaction(RT-PCR).Results: Lovastatin could inhibit the growth of the human colon carcinoma cell line HCT-116 and the inhibitory effect was dose-and-time dependent(P<0.01). Comparing with control group, the HCT-116 cell apoptosis was induced by Lovastatin at 48 hours,the cell apoptosis rate was gradually increased by lovastatin(p<0.05). Meanwhile, the ratio of G0/G1 phase was increased and the ratio of S phase was decreased significantly by Lovastatin(p<0.05). The expression of bcl-2 and MMP9 were downregulated and bax was upregulated by Lovastatin at 48 hours (p<0.01).Conclusion: Lovastatin could inhibit the growth of human colon cancer cell line HCT-116 in a dose-and-time dependent manner, and maybe its mechanisms were arresting cell cycle; inducing apoptosis through downregulating bcl-2 expression and upregulating bax expression . Relating to downregulating MMP9 expression.
