| Background and ObjectiveAngiogenesis is a hot spot in modern medical domain,in physiological conditions,angiogenesis not noly normally occurs during embryonic development, but also is the precondition of tissue and organ's differentiation.Unregulated angiogenesis is directly correlated with the development of mangy disease,such as cancer,psoriasis,irregular menses and arthritis and so on.That all are named angiogenesis diseases,which are shown the over or defect of angiogenesis.So the research about angiogenesis is very important.Angiogenesis is also crucial in the process of SA's synovium hyperplasia and pannus formation.SA belongs to the category of"arthralgia syndrome" in Traditional Chinese Medicine,which believes that impotency of Qi-blood circulation is the main pathology component,"blood stasis"is SA's crucial pathogenesis.In my experiment,SA rats' synoviums is my investigative target.Studying the relation of inflammation as well as angiogenesis and stagnation of blood,so as to explain that using huoxuehuayu-method cures SA rats' inflammation and angiogenesis,all these work mainly supplies theory demonstration about huoxuehuayu-method cures SA.Method1.The models of SA rats'angiogenesis were established,and observed the histopathological change.2.The feet swelling degrees of rats were measured,then calculate and contrast swelling rate.3.Study of analgesic effects of xuefuzhuyu-extractive through observing rats' stretching number and licking number.4.The level of serum NO was detected and the contents of serum TNF-αand IL-10 were determined by ELISA.5.Immunohistochemistry techniques have been used to detect cytokines VEGF and TGF-β1 in synovial tissue of SA rats.The above results were analyzed with image analysis system.Results1.The local joints of SA rats have obviously persistent swelling,many times of licking,and the colour pattern is eclipse,in the final stage,the local joints showed dim.The manifestations of synovial histopathology are inflammatory cell infiltration,lining cell proliferation,too much angiogenesis formation.2.Xuefuzhuyu-extractive can significantly decreased the swelling rate of joints of SA rats.3.Xuefuzhuyu-extractive can significantly inhibit the local pain of SA rats.4.Two weeks later of using xuefuzhuyu-extractive,the levels of administration team's serum TNF-αwas significantly decreased,serum IL-10 was significantly increased compared with control.The effect of serum NO had not statistical significance compared with control,but the tendency is decreased,on the whole, the best result was appeared in middle-dose team.5.According the result of synovium histochemical stain,xuefuzhuyu-extractive can effectively inhibit the express of VEGF and TGF-β1.Conclusion1.There are appearing a number of angiogenesis in the active phase of SA,the new vessels aggravate synovitis,which also result in the ruin of arthrodial cartilages and bones.2.Syndrome of blood stasis and SA have close relation,which is the one of pathogenesis of SA's active phase.3.Xuefuzhuyu-extractive can effectively improve the symptom of blood stasis, angiogenesis can be decreased by inhibiting the expression of VEGF and TGF-β1 in synovium. 4.The anti-inflammatory effect can be increased by adjusting the expression of serum TNF-α,IL-10 and NO.5.Xuefuzhuyu-extractive not only can improve microcirculation of the whole boby and the local joint so as to relieve pain,but also can inhibit the pathologic angiogenesis,which can prevent the destruction of joints,and improve the symptom of stagnation of blood.
