Objective: To observe the distribution changes of ICAM-1 VCAM-1 PECAM-1 and iNOS, and related mRNA among blood vessel endothelial cell in blood stasis rats; To explore the relationship between the adhesion molecules expression of vascular endothelial cell and TCM blood stasis by studying the effects of Xuefuzhuyu decoction on ICAM-1 VCAM-1 PECAM-1 and iNOS expression of vascular endothelial cell in rats with blood stasis.Methods: The models of blood stasis rats were prepared by injection of Adr. Sprague-daweley rats were randomized into five groups which consisted of control group, blood stasis model group, high doze Xuefuzhuyu decoction used group, moderate doze Xuefuzhuyu decoction used group and low doze Xuefuzhuyu decoction used group. Protein and mRNA expression of ICAM-1 VCAM-1 PECAM-1 and iNOS of vascular endothelial cell was assayed by immunohistochemistry in situ hybridization and RT-PCR.Results: (1)Protein and mRMA expression of ICAM-1 VCAM-1 PECAM-1 and iNOS of vascular endothelial cell in the models of blood stasis rats were increased comparing with control group. ICAM-1 expression of vascular endothelial cell in Sprague-daweley rats was increased obviously comparing with the control group (P<0.05). (2)VCAM-1 and PECAM-1 expression in the low doze Xuefuzhuyu decoction used group were markedly decreased comparing with the blood stasis model group. (3)When the rats with blood stasis were treated with Xuefuzhuyu decoction, protein and mRNA expression of vascular endothelial cell decreased markedly in dose-dependent. (4) VCAM-1 expression in the low and high doze Xuefuzhuyu decoction used groups were both markedly higher than control group.Conclusions: (1) Protein and mRMA expression of ICAM-1 VCAM-1 PECAM-1 and iNOS of vascular endothelial cell in the models of blood stasis rats were increasedcomparing with control group. 〥ifferent dozes Xuefuzhuyu decoction especially low doze group can decrease VCAM-1 and PECAM-1 expression, as well increase ICAM expression. (3)Different dozes Xuefuzhuyu decoction can distinctly decrease iNOS expression in blood stasis rats. 甀t might be one of the function of Xuefuzhuyu decoction to cure the blood stasis by decrease VCAM-1 PECAM-1 and iNOS expression of vascular endothelial cell.
