Objective:To study the effects of curcumin on the activity of ATPase and the mechanism of apoptosis in U-251 cell.Methods:1.U-251 cells were treated with 20, 40, 80, 100umol/Lcurcumin for 24h and the growth inhibition rates of U-251 cells were measured with MTT method.2.Cell apoptosis was inspected with flow cytometry(FCM).3. The activities of ATPase were determined by colorimetry method.Results:1.MTT assay: U-251 cells were treated with 20 ,4 0 ,8 0 ,1 00umol/Lcurcumin for 24h and the proliferation of U-251 cells were significantly inhibited in dose-dependant manner.compared with control group(8.42+2.57),growth inhibition rates of U-251 cells by curcumin had statistical significance. [(16.83+4.72 ) ,( 25.58+5.13), ( 43.50+6.09) and( 53.18+4.14)respectively].2. Flow cytometry : apoptosis of U-251 cells were induced significantly by the different concentration of Curcumin. Apoptosis rate of the groups were (16.47±4.72) , (25.62±5.13) , (40.80±6.09) and(54.92±4.14). compared with control group(3.56±2.57),there had statistical significance.3. colorimetry method: level of ATPase in U-251cells plasma membrane was low remarkably. Treated with 20, 40, 80, 100umol/L curcumin for 24h, the activities of Na+ -K+-ATPase of the groups were(1.34±0.41),(1.26±0.42),(0.60±0.09),(0.36±0.07)and(0.20±0.54)respectively;the activities of Ca2+-Mg2+-ATPase were ( 0.51±0.13 ),(0 .37±0.10),( 0.29±0.08),( 0.11±0.08 ) and( 0.05±0.19 ) respectively.the unit of ATPase is umolpi/ mgprot / hour。Excep -t that the groups of 0umol/L and 20 umol/L had on statistical significance.compared with the normal groups,the remaining groups of Na+ -K+-ATPase, there had statistical significance. Except that the groups of 20 umol/ and 40 umol/L,80 umol/L and 100 umol/L had on statistical significance,compared with the normal groups,the remaining groups had statistical significance.Conclusion:Curcumin induced apoptosis and inhibited proliferation via inhibition of activation of plasma Membrane ATPase.
