PART ONE EFFECT OF APIGENIN ON PROLIFERATION OF BREAST CANCER CELL LINE MDA-MB-231Objective: To investigate the effection of apigenin on proliferation of breast cancer cell line MDA-MB-231 in-vitro and explore the mechanism.Methods: MTT assay was used to examine the changes of cell proliferation. HE staining, microscopic observation and flow cytometry were used to detect the induction of apoptosis and the changes of cell cycle. Immunocytochemistry was performed to determine the protein of cyclin B1 and western blot was used to detect the protein of caspase-3.Results: (1)Apigenin significantly inhibit proliferation of MDA-MB-231 cells. Apigenin and inhibition between time-and does-dependent manner.(2) In result of TEM, after dealt with 38.1μmol/L for 48 hours, microvillus disappear, electron density of cytoplasm is high, large deciduous apoptosis bodies appear. (3)In result of FCM after dealt with 40,80μmol/L for 48 hours, cell cycle is controlled in G2/M phase and there is a apoptosis peak in DNA trapes.(4)In result of immunohistochemistry, the protein level of cyclin B1 was gradually downregulated with concentration increasing of apigenin.(5)Western blot results show that apigenin induced apoptosis in increasing the protein levels of caspase-3.Conclusion: The anti-tumor mechanism of apigenin is that it can significant inhibits the proliferation of human breast cell line MDA-MB-231, mainly controlls cells in G2/M phase and increases the expression of caspase-3 to induct apoptosis.PART TWO EFFECT OF APIGENIN ON INVASION AND METASTASIS ABILITIES OF BREAST CANCER CELL LINE MDA-MB-231Objective: To investigate the effection of apigenin on invasion and migration of breast cancer cell line MDA-MB-231 in-vitro and explore the mechanism.Methods: MTT assay was used to test the adhesion of MDA-MB-231 cells. Millicell chambers were utilized to test the ability of the invasion and migration of MDA-MB-231 cells. Immunocytochemistry was performed to detect the protein of uPA and MMP-9.Western blot was used to determine the protein of p38MAPK.Results: (1)Apigenin inhibit the invasion and migration of MDA-MB-231 cells in does-dependent manner.(2)In result of immunocytochemistry the expressions of uPA and MMP-9 were gradually downregulated with concentration increasing of apigenin.(3)Western blot results show apigenin decreased the protein level of p38MAPK.Conclusion: Apigenin inhibits the adhesion, invasion and migration of human breast cell line MDA-MB-231. This may be associated that apigenin decreasd the expressions of uPA and MMP-9 through inhibiting p38MAPK signal conduction path.
