Objective: To investigate the expressions and significance of Bcl-2,Caspase-3 in tissues of breast cancer patients with bone metastasis,analyze the relationships between their expressions with the clinical pathological factors,and approach the risk factors of bone metastasis.Methods: Immunohistochemistry method was used to measure the expressions of Bcl-2 and Caspase-3 in 40 breast cancer patients with bone metastasis,30 breast cancer patients without bone metastasis and 30 benign breast hyperplasia tissues respectively.The bone metastasis characteristics diagnosed by Whole Body Bone Imaging(WBI)were analyzed.Moreover,the correlation between bone metastasis and the expressions of Bcl-2,Caspase-3 and the relationship between these two proteins expressions with the clinical pathological factors of breast cancer patients with bone metastasis were also analyzed.Results1.The bone metastasis of breast cancer invaded in spine accounts for 60%,higher than that in chest(57.5%)? pelvis(42.5%)? limb(20%)and skull(7.5%)respectively,the statistical difference was significant(?2 = 36.380,P<0.01).2.The positive expression rate of Bcl-2 in 70 patients with breast cancer was 71.4%,significantly higher than that in 30 benign breast hyperplasia tissues(46.7%),the statistical difference was significant(?2 = 5.589,P<0.05).Bcl-2 positive expression rate in 40 breast cancer patients with bone metastasis was 72.5%,slightly higher than that in 30 patients without bone metastasis(70%),which had no statistically significant difference(?2 = 0.053,P>0.05).3.The positive expression rate of Caspase-3 in 70 patients with breast cancer was 82.6%,slightly higher than that in 30 patients with benign breast hyperplasia tissues(80%),without significant statistical difference(?2 = 0.116,P>0.05).The positive expression rate of Caspase-3 in 40 breast cancer patients with bone metastasis was 92.5%,which was significantly higher than that in 30 patients without bone metastasis(70%),and had statistically significant(?2 = 6.11,P<0.05).4.The positive expression rate of Bcl-2 in breast cancer patients with bone metastasis was related to the expressions of ER and PR(?21= 7.595,P1<0.01;?22= 5.021,P2<0.05),and there was a remarkably positive correlation(r1= 0.498,P1<0.01;r2= 0.411,P2<0.01)between their expressions.But,the positive expression rate of Bcl-2 was not related to other clinical pathological factors including age?tumor size?histological grade?axillary lymph node metastasis?the expressions of HER-2 and Ki-67(P>0.05).5.The positive expression rate of Caspase-3 in breast cancer patients with bone metastasis was not related to all clinical pathological factors,including age?tumor size?histological grade?axillary lymph node metastasis?the expressions of HER-2,ER,PR and Ki-67(P>0.05).6.There was no correlation between the expressions of Bcl-2 and Caspase-3 in the breast cancer tissues(P>0.05).7.The bone metastasis from breast cancer was related to the positive expression of Caspase-3.The bone metastasis rate in the positive group of Caspase-3 was 62.7%,significantly higher than that in the negative group(27.2%),having statistically significant difference(?2 = 4.755,P<0.05).The bone metastasis from breast cancer was also related to the axillary lymph node metastasis.The bone metastasis rate in the group of axillary lymph node metastasis was 69.7%,significantly higher than that in the group of non-axillary lymph node metastasis(45.9%),showing significantly statistical difference(?2 = 4.018,P<0.05).The bone metastasis from breast cancer was not correlated with other factors including age?tumor size?histological grade?the expressions of Bcl-2,ER,PR,HER-2 and Ki-67(P>0.05).Conclusions1.Bcl-2 and Caspase-3 are highly expressed in breast cancer tissues,but there is no correlation between the expressions of these two proteins,suggesting that the apoptosis pathways mediated by Bcl-2 and Caspase-3 may play an important role in the occurrence and development of breast cancer,which may provide new targets for the diagnosis and treatment of breast cancer.2.There is no difference in the expressions of Bcl-2 in breast cancer with or without bone metastasis.But,it is a positively correlated with the expressions of ER and PR,suggesting that the expression of Bcl-2 may be one of the bases for endocrine therapy in breast cancer patients with bone metastasis.3.The expressions of Caspase-3 are significantly different between breast cancer with and without bone metastasis,suggesting that the high expression of Caspase-3 is related to bone metastasis in breast cancer,and just like axillary lymph node metastasis,it may be regarded as a risk factor of bone metastasis in breast cancer.
