Propofol, is commonly used in clinic intravenous anesthesia, and its chemical name is 2,6-di(2-methylethyl)phenol. It is widely used on induce and sustain in anesthetic operation, which is belonged to center anesthesia. The anesthetic mechanism of propofol is related with endogenous inhibitor GABA(Gamma aminobutyl acid) and the structure of its receptor, on which mainly enhanced biological inhibition of GABA. Propofol has many advantages than other anesthetic drugs. In this paper, it designs a new prodrug of propofol withγ-amino butyl acids and synthesized by new methods, with the knowledge based on the physicochemical property and the principles of prodrugs. The structure of the prodrug is also confirmed. Unexpectedly,after the determination experiment on drug action of the new drug, it is concluded that this prodrug water-solubility is worse than before and it has not on rats. The failure case can be a research reference in the future.
