The dry rhizome of Iris tectorum Maxim. (Iridaceae) can be used to removing food retention, breaking blood stasis, excreting water, and detoxifying. It is widely distributed in guangdong, guangxi, sichuan and guizhou province.This work includes two areas. First, a systemic study on the chemical constituents in the rhizome of Iris tectorum Maxim. was made by kinds of separation methods and structure identification methods. Then four active constituents in rhizome of Iris tectorum Maxim. and Belamcanda chinensis(L.)DC. were analyzed and compared.Chemical study on Iris tectorum Maxim. The ethyl alcohol extracts of rhizome of Iris tectorum Maxim. were extracted by petroleum ether, ethyl acetate and n-Butanol by turns. Each part was separated and purified using the silica gel column, Sephadex LH-20 and crystallization. Eleven compounds were isolated and all of them were identified by ESI-MS, 1H-NMR and 13C-NMR as: tetradecanoicacid(A1), dihydro-kaempferide(B1), 5,7,4′-trihydroxy-6,3′-dimethoxy isoflvone(B2), apocynin(B3), iristectorigenin A(B4), (1,1′-(6,6′-Dihydroxy-5,5′-dimethoxy [1,1′-biphenyl]-3,3′-diyl)bisethanone)(B5), iristectorin B(B6), tectorigenin(C1), iristectorin A(C2), tectoridin(C3), tectorusid(C4). Compounds A1 and B2 were isolated from this plant for the first time. Compound B5 was isolated from the natural plants for the first time.Determination of isoflavone content in Iris tectorum Maxim. and comparative study of Belamcanda chinensis(L.)DC. In order to evaluate the isoflavonoids in rhizome of Iris tectorum Maxim. and Belamcanda chinensis(L.)DC., HPLC method was applied to determine the contents of active constituents in both of them. The content was analyed by external standard method. The results showed that chemical composition of them were very similar and contents of four isoflavonoids in rhizome of Iris tectorum Maxim. were higher. It is hinted that the rhizome of Iris tectorum Maxim. may have the same effectiveness with Belamcanda chinensis(L.)DC. in some areas and it may even have a better effect. The chemical constituents of four isoflavones content in Iris tectorum Maxim. were: tectoridin 4.30%, iristectorin B 0.35%, tectorigenin 1.45%, 5,7,4′-trihydroxy-6,3′-dimethoxy isoflvone 0.24%, it can be used to develop formulations of isoflavonoids.
