Objective:To study the proliferation and ostogenic differentiation effect of different concentrations of dexathasone on adipose tissue-derived cells of rat in virro.Methods:The adipose tissue were isolated from the inguinal fat pads of the SD rats,then placed in Dulbeco Modified Eagle Medium(EMDM) with fetal bovine serum in primary cultivation.primary ADSCs were obtained by the methods of college-nase I digestion.subsequently,the ADSCs were cultured and subcultured.cell morphology and prociferation characteristic were observed under inverted mircroscope,light mircroscope.The third of ADSCs were induced into osteo-blasts by the osteogenic inducing fluid including with dexamethasone,ascorbic acid,beta-sodium glycerphosphate.The osteogenic phenotype were examinated by means of MTT method,alkaline phosphatase(ALP),Von Kossa stainings and immunocytochemisty.In subcultures.MDSCs were respectively treated with different concentration dexamethasone (10-8mol/L,10-7mol/L,10-6mol/L and 10-5mol/L).Results:ADSCs were succeefully obtained and cultured from rat adipose tissue.They appeared rapidly.After include with dexamethasone,ascorbic acid,beta-sodium glycerphosphate.ADSCs expressed the characteristic of osteoblasts:increasing of alkaline phosphatase(ALP) actiuity signifinally appearance of calcium node showed under Kossa staining and collagen I through immunocytochemistry.While those of the control group were negative. Dexamethasone of 10-8 mol/L concentration has not inhibitory efect on growth and proliferation of adipose-derived stromal stem cells(P> 0.05).Dexamethasone of over 10-7 mol/L concentration has inhibitory efect on growth and proliferation of adipose-derived stromal stem cells (P<0.05).Conclusions:Dexamethasone of over 10-7 mol/L concentration has inhibitory efect on growth and proliferation of adipose-derived stromal stem cells.Under the same jnduction condition.10-7mol / L dexamethasone is efective to differentiate into osteoblast from adipose-derived stromal stem cells.
