ObjectiveTo study and prepare osteogenic-activitic bionic periosteum implanted bone defects areas to repair it,providing new ideas to address the bone defect , then for clinical practice, and providing experimental basis to ultimately replace the autogenous bone and allogenic bone graft in bone defect.MethodsUsing the technology and methods of tissue engineering, applying the combining method of histology immunohistochemistry and animal experiment for the experimental study. Selecting adipose-derived stem cells(ADSCs) as seed cells, osteoblasts revulison to osteogenic cells in vitro,were detected by cell morphology, and were identified to osteogenic differentiation by biological characteristics,alkaline phosphatase, alizarin red staining, von Kossa staining techniques indicators. Implanting bionic periosteum of osteogenic cells as a scaffold material with small intestinal submucosa (SIS) cultured construct complex functional cells - composite body in vitro to subcutaneously of nude mice, observating the bone formation and their cellular compatibility,by histology, transmission electron microscopy immunohistochemical. Finally, building a bone defect animal model, then implanting periosteum bionic to bone defects to repair it, through X-ray, and other organizations, identifing the morphological detection mechanism and the similarity with autologous bone graft into the bone periosteum , determining the biomimetic periosteum of repairing bone defects.ResultsSeparating and amplificating rabbit adipose-derived stem cells In vitro, by induction, expressing the osteogenic bone cell phenotype. Combing osteogenic adipose-derived stem cells with SIS may build a bionic periosteum with physical structure and function,and implante it to nude mice can form new bone tissues with good blood supply . Bionic periosteum repairing rabbit segmental bone defects can form good effect, and its bone mechanism was similar to the autologous periosteum graft into the bone.ConclusionAdipose-derived stem cells can transforme to osteogenic bone cells by osteogenic culturing, can express the phenotype of bone cells and provide an experimental basis of adipose-derived stem cells as seed cells for bone tissue engineering. The effect of bionic periosteal which was build by SIS and adipose-derived stem cells in animal experiments, determining the SIS as a scaffold material can provide a good metabolism environment of the tissue engineering bone , suggesting a good biocompatibility. The experimental research of bionic periosteal repairing long bone defect in rabbit , made it to be an effective means of tissue engineering periosteum repairing large bone defect.SignificanceIn recent years, the rapid development of tissue engineering techniques , made modern medicines were out of the scope of organization, organ transplantation, into a new era of tissues,and organs recycling.Bone defects was common orthopedic disease in clinical ,the repairing of bone defects was one of the clinical problems. Using bone tissue engineering technology to produce bionic periosteum for bone repair ,providing a new ideato solve this problem, hoping use the patient's own adipose-derived stem cellswith SIS can build biomimetic periosteum used in clinical bone defects and bone nonunion treatment, broughting innovative changes to the treatment of bone defects .
