【Objective】To observe the expressions of vascular endothelial growth factor(VEGF),erytbxopoietin(EPO),erythropoitin receptor(EPOR) in different levels of glucose in the retinal Mtiller cells of mice.So as to investigate the role of M(u|¨)ller cells in the progress of diabetic retinopathy.【Methods】(1) Adopt enzyme digestion to culture the M(u|¨)ller cells of mouse at postnatal 7 day.(2) Divided into 4 groups accordding to the concentrations of glucose in DMEM:25mmol/L,30mmol/L,40mmol/L, 50mmol/L,and the 25mmol/L group was as the control group.Cultured for 1 day,3 days,5 days.(3)The expressions of VEGF mRNA,EPO mRNA,EPOR mRNA were detected by RT-PCR.【Results】The expressions of VEGF mRNA,EPO mRNA,EPOR mRNA in retinal M(u|¨)ller cells were significantly enhanced in the high glucose group which increased obviously(P<0.05),but the difference between 50mmol/L group and the 40mmol/L group did not have statistic significance(P>0.05).【Conclusion】(1)With glucose concentration elevating,VEGF expression of cultured retinal M(u|¨)ller cells increased.(2) With glucose concentration elevating,EPO,EPOR expression of cultured retinal M(u|¨)ller cells increased.Therefore,the upregulation of the expression of VEGF,EPO,EPOR in the retinal M(u|¨)ller cells in DM patients may be contribute to diabetic retinopathy.
