Objective:Through exploring the expression of matrix metalloproteinase-9(MMP-9) and urokinase-type plasminogen activator(uPA) in WHO gradeⅠmeningiomas(from WHO Year 2000 Classification),to further investigate the relationship between the action of MMP-9 and uPA and the invasiveness of WHO gradeⅠmeningioma, and offers theoretical and experimental evidence for studying WHO gradeⅠmeningioma.Methods:The expression of MMP-9 and uPA was determined by immunohistochemistry in 50 cases of human WHO gradeⅠmeningiomas and 10 normal human meninges by random sampling,including invasive meningioma 18 cases,noninvasive meningiomas 32 cases.Use multiple functional true-color pathological analyzing system to analyze the results, and use statistics to test their validity.Results:The invasive group had a significantly higher expression of MMP-9 than that of noninvasive group(x~2=4.608,P<0.05).The invasive group had a significantly higher expression of uPA than that of noninvasive group(x~2=4.151,P<0.05).Expression of MMP-9 and uPA was negative in 10 samples of normal human meninges tissues.Moreover, the expression of MMP-9 was significantly positive correlated with uPA (correlation coefficient r=0.679,P<0.05).Conclusions:1.The expression of MMP-9 in meningiomas were higher than those in normal meninges;and the expression of MMP-9 in invasive meningiomas were higher than those in noninvasive meningiomas.In invasive meningiomas it was higher than that in noninvasive meningiomas.This indicated that MMP-9 played an important role in the course of invasion of meningiomas.2.The expression of uPA in meningiomas was higher than those in normal meninges;and the expression of uPA in invasive meningiomas were higher than those in noninvasive meningiomas.In invasive meningiomas it was higher than that in noninvasive meningiomas.The uPA protein accumulation correlated with the development and the invasiveness of meningiomas.3.There is a significant positive correlation between the expression of MMP-9 and uPA.Significant correlation was observed between high-expression of MMP-9 and high-expression of uPA with the invasiveness of the meningiomas.This suggested that there may be a mutual synergy between MMP-9 and uPA in the occurrence and development of WHO gradeⅠmeningiomas.This study demonstrates the relationship between MMP-9 and the invasiveness of WHO gradeⅠmeningiomas,and the relationship between uPA and the invasiveness of WHO gradeⅠmeningiomas.They have significance to evaluate the invasiveness biological behavior of meningioma,and can provide evidence for instituting reasonable therapy scheme,judging prognosis and assessing curative effect in clinic.
