ObjectiveExamining the body weight, blood sugar of low birth weight rats after given L-Arg, observing the PI3K and PKB in local hepatic tissue. Discussing the possible mechanism of improvinging insulin resistance by using L-Arg.Methods1. Establishment the models of low birth weight rats: Eighteen male and eighteen female healthy three-month SD rats were bought. Male and female rats were put into a cage at the proportion of 1:1. The pregnant rats were divided into three groups: pregnant rats in model group and intervention group were fed with a 10% low protein diet, while normal group were fed with a 21% normal protein diet. 21% normal protein diet was given to the maternal rats during lactation and the offspring after ablactation. The maternal rats of intervention group were given a supplementation of L-Arg drinking water during the peroid of lactation.2.Body weight, blood sugar of the offspring were measured at the age of 1w, 3w and 8w . 3. Measurement of important factors of PI3K pathway: Expressions of the protein of PI3K and PKB were detected by western-blot at the age of 1w, 3w and 8w.4. Statistical analysis: All date were statisticed by the SPSS 13.0. Difference between groupse were evaluated using method in One-Way ANOVA , rate comparison between groups were evaluated using t test. Significance was set at P <0.05.Results1. Establish low birth weight rat model: The offspring in model and intervention groups got low birth weight. But there were no obvious changes in the newborn member per nest and perinatal death rate among three groups.2. At the age of 1 w, offspring's body weight in model or intervention group was lower than that of normal group, but the intervention group was heavier than the model group. At the age of 3w, the body weight in model group was lower than that of normal group, the body weight in intervention group which is close to that of normal group is higher than that of model grouop. While at the age of 8w, the body weight in model grouop was lower than thar of normal group. There was no difference between intervention group and normal or model groups. Intervention group was heavier than model group, but without significant difference.3. The difference of blood sugar among three groups was not obvious at the age of 1w, 3w and 8w.4. At the age of 1w, 3w and 8w, expression of PI3K in model group was lower than that of normal group. At the age of 1w, expression of PI3K in intervention group was higher than that of model group, but without significant difference when compared to normal group. At the age of 3w and 8w, expression of PI3K in the intervention group was between normal group and model group, but without significant difference when compared to normal or model group. As for expression of PKB at the age of 1w and 3w, there was no significant difference among three groups. At the age of 8w, expression of PKB in model group was lower than that of normal group, but it was was higher in intervention group than that of model group, and without significant difference when compared to normal group. but it was higher than that of model group, and there was no significant difference when compared to normal groupConclusion1. The low expression of PI3K and PKB is one of the main mechanisms of hepatic insulin resistance.2. Supplement of L-Arg in the early life could promote development and catch-up growth in low birth weight rats.3. Supplement of L-Arg in the early life could increase expression of PI3K and PKB to improve hepatic insulin pathway and improve hepatic insulin resistance.
