Objective:To explore the further pathogenesis of GBS and provide the theory of a new potential therapy method for GBS and other autoimmune disease,The expression of CD22 and CD72 of B lymphocyte and CD72 mRNA in peripheral blood mononuclear cells(PBMNCs) of GBS were tested.Methods:36 patients with GBS and 20 healthy controls(HC) were investigated.The expression of CD22 and CD72 on peirpheral B lymphocytes were examined by flow cytometry.The expression of CD72 mRNA in peripheral blood mononuclear cells was detected by RT-PCR.Furthermore,the corelation between the expression of CD22 or CD72 and the severity of the disease were analyzed.Results:(1) In GBS group,total B lymphocyte numbers was 606±118,The CD22~+CD19~+ and CD72~+CD19~+ were 78.73±7.43 and 62.11±9.14 respectively.In normal control group,B lymphocyte numbers was 248±92,The CD22~+CD19~+ and CD72~+CD19~+ were 83.38±9.46 and 69.72±11.42 respectively.Compared with normal control group,B lymphocyte numbers were significantly increased(P<0.01),the CD72~+CD19~+decreased significantly(P<0.01)and the CD22~+CD19~+ was of no significant differences(P>0.05) in GBS group.(2) In the acute stage group,B lymphocyte numbers was 682±91,The CD22~+CD19~+ and CD72~+CD19~+ were 77.99±8.10 and 57.79±7.69 respectively.In recovery stage group,B lymphocyte numbers was 550±111,the CD22~+CD19~+ and CD72~+CD19~+ were 79.75±6.50 and 68.14±7.58 respectively.Compared with recovery stage group,B lymphocyte numbers were significantly increased(P<0.01),the CD72~+CD19~+ decreased significantly(P<0.01)and the CD22~+CD19~+ was of no significant differences(P>0.05) in the acute stage group.(3)In mild group, B lymphocyte numbers was 561±96,The CD22~+CD19~+ and CD72~+CD19~+ were 78.38±8.64 and 65.27±8.46 respectively.In severe group,B lymphocyte numbers was 685±116,The CD22~+CD19~+and CD72~+CD19~+ were 79.33±4.85 and 56.41±7.65 respectively. Compared with mild group,B lymphocyte numbers were significantly increased(P<0.01),the CD22~+CD19~+ and CD72~+CD19~+were of no significant differences(P>0.05) in severe group.(4) The expression of CD72 mRNA in PBMNCs was 0.21±0.07 in GBS group,0.35±0.06 in healthy controls.Compared with healthy controls,CD72 mRNA Was decreased in GBS group(P<0.01).(5)The expression of CD72 mRNA in PBMNCs was 0.18±0.03 in the acute stage group,0.26±0.04 in recovery stage group.CD72 mRNA was decreased in the acute stage group,compared with recovery stage group(P<0.01).(6)The expression of CD72 mRNA in PBMNCs was 0.24±0.04 in mild group,0.19±0.02 in severe group.CD72 mRNA was of no significant differences in mild group,compared with severe group(P>0.05).Conclusions:1.Inhibitory co-receptor CD22 in the pathogenesis of GBS may not be significant.2.Inhibitory co-receptor CD72 lower expression may decreases the threshold of B cell activation and thus affects B cell function leading to GBS.
