Objective:Observe the effect of Jianpi Shugan Huoxue Fang (JPSGHXF) on IL-2 and TGF-β1 of Serum. Discussion the changes of tumor PTENgenes expression. Research the mechanism of action of JPSGHXF.Methods:We built animal model of H22 hepatic cancer with mice according to inoculate tumor cell.Mice were randomly divided into six groups such as the normal group,the model group,CTX group,low-dose, middle-dose and high-dose group of JPSGHXF.Observe the mices'overall condition.The mices were executed at the 15th day after being treated with medicine.Then use the mices'serum,thymus,spleen and cancerous tissue to observe the thymus index, spleen index and inhibitory rate.We detect the content of IL-2 and TGF-(31 in serum with the method of enzyme linked immunosorbent assay(ELISA). The gene expression of JPSGHXF on PTEN mRNA of cancerous tissue was examined with reverse-transcription polymerase chain reaction(RT-PCR).Results:①The animal model of Hepatocarcinoma H22 Tumor-bearing Mice was set up successfully.②JPSGHXF could improve the whole condition of tumor-bearing mice, and enhance the thymus index and spleen index.③The middle-dosee and high-dose of JPSGHXF had better inhibitory rate of tumor cells,which inhibitory rate were 47.69% and 35.56%.④The middle-dose of JPSGHXF could raise the content of IL-2 in serum, The low-dose and middle-dose of JPSGHXF had the best effect on IL-2 (P<0.05),But,no better than CTX.⑤The low-dose.middle-dose and high-dose of JPSGHXF could raise the content of TGF-β1 in serum, and had significantly statistical meaning in comparison with model group (P<0.05).⑥JPSGHXF groups could reduce the expression of PTEN mRNA. Compared with the model group, the middle-dose were significantly effective and had significantly statistical meaning (P<0.05).Conclusions:JPSGHXF had obvious depressant effect on H22 hepatic cancer mice.The molecular mechanism may related to such factors as below:①raising the content of IL-6 and TNF-αin serum to strengthen immune function②decreasing expression of TGF-β1 mRNA of the cancerous tissue to strengthen immune surveillance function. Moreover, the experiment proved that the antitumous effect of JPSGHXF is related with dosage of medicine.
