Objects:Recent studies into the pathogenesis of asthma have revealed a dynamic role for Th17 cells in the perpetuation of airway inflammation via secretion of IL-17.Because IL-17 production is restricted to Th17 cells in CD4~+T cells,the effects exerted by IL-17 may help one to understand the contribution of Th17 cells to the inflammatory response.In this study,we evaluated whether IL-17 could enhance IL-8 release from human peripheral blood CD4~+T cells.Methods:Five healthy volunteers and ten acute attack asthma patients were collected from the out-patient clinic of the second hospital of central south university.The CD4~+T cells which obtained from peripheral blood of all subjects with magnetic separation were divided into four groups with the same cell concentration of 2×10~6/ml,which co-cultured respectively with hIL-17,hIL-17+Anti-hIL-17Ab,hIL-17+IgG and control.36 hours later,we get the supernatant of cocultured human CD4~+T cell and detect the concentration of IL-8 using enzyme linked immunosorbent assay(ELISA).Results:1.The purity of peripheral blood CD4~+T cell using magnetic separation is very high(97.3%).2.The supernatant IL-8 concentrations of CD4~+T lymphocytes derived from acute attack asthma patients were higher than that of the healthy volunteers.3.The supernatant IL-8 concentrations of the groups which co-cultured with Anti-hIL-17Ab were decreased significantly than that of other groups.Conclusions:We demonstrate IL-17 enhancd IL-8 release from human peripheral blood cells in acute attack asthma.Th17 cell possibly undergoing IL-17 play an important role in the pathogenesy of acute attack and exacerbation asthma.
