Background Chromosome instability is considered characteristic of malignant cell changes,and abnormal mitotic cells can cause chromosome instability. Centrosome as the cell's main microtubule organizing center,set up mitotic spindle and regulating mitosis in the cell cycle,,thereby plays an important role in maintaining the stability of the chromosome.Therefore,tumor cell gene instability and centrosome abnormalities are closely related to.Been found with a lot of centrosome abnormalities closely related gene,serine / threonine kinase 15 (STK15) gene that is one of its encoding a centrosome replication-associated kinase,and has a high level of tyrosine kinase activity involved in the regulation of centrosome maturation,G2 / M phase conversion,set up the bipolar spindle and chromosome separation and so on mitotic case.Recent studies show that the serine / threonine kinase 15 in a variety of malignant tumors in the over-expression,leading to abnormal centrosome amplification,aneuploidy formation and neoplastic transformation and the biological behavior of tumor-related.Its expression in the non-hodgkin's lymphoma study reported less domestic. OBJECTIVE Immunohistochemical SP staining was adopted to detect the expression of STK15 in 42 cases of surgical excision of NHL and 12 cases of reactive hyperplasia in lymph node tissue samples,to explore the role of of serine /threonine kinase 15 in occurrence and development of NHL and its relationship with clinicopathological features.METHODS NHL diagnosed in 42 cases of surgical resection in patients with lymphoid tissue and 12 cases of lymph node reactive hyperplasia in patients with surgical resection of lymphoid tissue was collected as the study object,using immunohistochemical SP staining to detect the expression of STK15 gene,using Leica image analysis software IM50 calculated the percentage of positive area,we did the statistics analysis with chi-square test and t test methods.RESULTS1.STK15 gene was not expressed in lymph node reactive hyperplasia.2.The expression rates of STK15 gene in NHL was 86%(36/42),including follicular lymphoma 62.5%(5/8),diffuse large B-cell lymphoma 92.3%(12/13), angio-immunoblastic T-cell lymphoma 85.7%(6/7),anaplastic large cell lymphoma 100%(5/5),mantle cell lymphoma 80%(4/5),peripheral T cells lymphoma 100%(4/4).The expression rates of STK15 gene in NHL significantly differenced from which in lymph node reactive hyperplasia(P<0.05).3.WHO2000 reference standard will be phased NHL specimens,one ofⅠperiod of four cases,Ⅱperiod of 16 cases,Ⅲperiod 18 cases,ⅣPhase Example 4,after statistical analysis,The expression rates of STK15 gene at each period of NHL has significant difference(P<0.05).4.The expression rates of STK15 gene in indolent lymphoma groups significantly differenced from which in aggressive lymphoma groups(P<0.05).5.There was no correlation between the expression of STK15 gene with age, sex,the scope of extra-nodal disease,LDH,IPI.CONCLUSIONS1.The expression rates of STK15 gene in NHL significantly differenced from which in lymph node reactive hyperplasia,so to detect the expression of STK15 gene have clinical significance to identification the two diseases.2.The expression rates of STK15 gene at each period of NHL has significant difference,so to detect the expression of STK15 gene have guiding role to determine the clinical stage of NHL.3.The expression rates of STK15 gene in indolent lymphoma groups significantly differenced from which in aggressive lymphoma groups,so to detect the expression of STK15 gene have clinical value to estimate the malignant degree of lymphoma.4.At present,three types of STK15 gene kinase inhibitors have been found, and in vitro experiments have confirmed its role to inhibit the role of STK15 gene, by further in-depth study on the STK15 gene targeted therapy will likely become an effective means of treatment of NHL.
