Background and Objectives Epilepsy is the familiar nervous system diseases in children,has high incidence.When dignosis epilepsy,we must hold antiepileptic drugs(AEDs) for long times to control convulsion,disense brain injure,improve prognosis,increase life quality.Thyroid gland is the important incretion gland in human, the most physiology function of thyroid hormone is to advance energy and matter metabolice,advenca grow and development process.If shortage of thyroid hormone in childhood can effect nervous system's development,cause brain develop obstacle,effect frame and intelligent development.Long-times use AEDs can effect liver,kidney,blood, bone,female reproduce,has few article of effection on thyroid hormone and struction, above all new type AEDs,for example oxcarbazepine and levetiracetam,have no article of the effection on thyroid hormone.For this purpose,we plan this study to evalutiong the impact of thyroid structure and endocrine function based on anti-epileptic drugs of monotherapy and united drug.Methods A total of 60 health female Wistar rats(age 21~22days,weight 48~58g),which were randomly divided as valproate acid(VPA) group,oxcarbazepine(OXC) group,levetiracetam(LEV) group,VPA+LEV group,OXC+LEV group,control(CON) group.The weight of rats in different groups have no statistice difference.Respectively feed VPA 200mg/kg.d bid;OXC 100 mg/kg.d bid;LEV 100 mg/kg.d bid;VPA 200mg/kg.d bid+LEV 100 mg/kg.d bid;OXC 100 mg/kg.d bid+LEV 100 mg/kg.d bid; control group feed same dosage physiology brine.Weight per 7 days,feed drug in accordance with weight.We feed these rats with drugs twice daily in accordance with the grouping,which are Sustained 90 days.Blooding,putting to death,Separatiing of thyroid organization,using RIA to determine serum TT3,TT4,TSH level.We use HE to stain thyroid tissues.Results1.Thyroid hormone:When Compared with the reference group,VPA group TSH decrease(P<0.01 ),TT3,TT4 have no statistically significant correlations(P>0.05). OXC group TT3,TT4 decrease(P<0.01 ),TSH increases(P<0.05);LEV group TT3,TT4,TSH have no statistically significant correlations(P>0.05);OXC+LEV group,VPA+LEV group TT3,TT4,TSH have no statistically significant correlations (P>0.05).2.Thyroid structure:VPA group thyroid follicular volume slightly reduced;OXC group follicular volume Increased,epithelial cells thinning;LEV group have no effection, OXC+LEV group与VPA+LEV group follicular volume size different.Conclusions1.VPA can reduce TSH,long-term application may cause disorders of thyroid structure and hormone secretion.2.OXC can reduce TT3,TT4 and increase TSH,follicular volume Increased,epithelial cells thinning,effect thyroid function.3.LEV have no effection on thyroid hormone and thyroid structure,with VPA and OXC united can reduce their effection on thyroid gland.4.We must monitor thyroid function regular when we use AEDs for long time.
