PARTI THE MORTALITY RATES AND TREND OF LEUKEMIA DURING 1970-2005 IN SHANDONG PROVINCEObjective To analyze the mortality and trend of leukemia from 1970 to 2005 in Shandong province, and to provide clues of causes, treatment and prognosis of leukemia.Methods Data of death retrospective survey of all causes during 1970-1974, 1985-1989, 1990-1992, and 2004-2005 had been selected in Shandong province. Leukemia death rates and standardized mortality rates by the percentages of world population were calculated. The trend was analyzed using Joinpoint regression model.Results The mortality rates of leukemia in the four periods were 1.97, 3. 63, 3.96, and 4.36; the rates for male were 2.20,4.05,4.25, and 4.86; and the rates for female were 173, 3.19, 3.66, and 3.85 per 100,000 persons respectively. In the specific-age mortality analysis there was a small peak of death rates in less than 5 years old groups, and the slop of death rates increased from 60 years old and reached high peak in 80 years old. The trend of mortality analysis showed an increasing trend during 1970-2005. After correction missing data, there was 3.546 percent average annual increase, a statistically significant, in the period time of from 1970 to 1992, and 0.445 percent after 1992. In the sample survey counties, we could find that the leukemia mortality rate increased greater in industrial relatively faster economic development area.Conclusions There was a slowly increasing trend of the mortality of leukemia during the past 35 years in Shandong province. PART II THE EXPERIMENTAL STUDY OF ANTI-TUMOR EFFECTS OF GARLICINObjective To investigate the inhibitory effect of diallyl disuIfide(DD) and diallyl trisulfide(DT), the two main components of garlicin, respectively, and combined treatment of DD and DT with adriamycin(ADM) on cell K562/AO2 and K562/AO2, which indicate that DD and DT can reverse drug resistance.Methods After treatment with DD and DT for different time periods(24h, 48h, 72h), the viability of the cells was determined by trypan blue staining. The 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyltetrazoliumbromide(MTT) assay was used to survey the proliferation of K562 and K562/AO2 cells treated by DD and DT respectively and combined with ADM.Results Trypan blue staining proves that low concentration of DD and DT have little significant side effects on the cells. Compared with the control group, the DD and DT group, including K562 and K562/AO2 cells, are down to varying degrees, and the inhibitory rates increase along the concentration of the drug, which means in a dose-effect relationship. DD and DT combined with ADM on cell K562/AO2 respectively, and the IC50 of ADM significantly decrease, which shows the role of reversible resistance to ADM of DD and DT.Conclusions DD and DT both can inhibit the cell proliferation respectively. Furthermore, DD and DT can enhance the inhibition of ADM on cell K562/AO2, reducing the amount of ADM, having statistical significance on the reversal of resistance.
