As the pollution and global warming became more and more serious, cyanobacteria occurs more frequently and extensively recently. Microcystins(MCs) were produced by many cyanobacterials. They threatened drinking-water and aquatic animals. Monitoring of microcystins (MCs) in fish and other aquatic animals is important for evaluating the potential risk for human consumption. This experiment tried to establish a method to determine MCs in aquatic products rapidly and widely. The research contained following aspects.1. A comfirmative method was developed with high performance liquid chromatography to detect MC in aquatic products. The conditions of chromatography was optimized by examining column, temperature and mobile phase composition. Diode array detector was used at 238nm wavelength test. The detection limit of MC was 33.3ng/g. This analytical method presented a simple and rapid procedure for determining MC, and could apply to most of aquatic products.2. The condition of primary treatment such as extraction method, solid-phase extraction columns, eluate and eluent were optimized. The treatment involved extraction of the analytes with 85% methanol and clean-up by SPE with a Envi C18 cartridge. Several aquatic products were tested with HPLC. The recovery of spiked samples was well, and confirmed with LC/MS. The recoveries of MC-RR addition were from 70.5% to 85.8% and the relative deviation was below 5.6%. The recoveries of MC-LR addition were from 69.3% to 84.7% and the relative deviation was below 5.5%.3. This analytical method presented a simple and rapid procedure for determining MC, and could apply to most of aquatic products. In addition, it tried a new method to detect toxicity of MC in water and aquatic products by light-emitting bacteria.
