Molecular imprinting polymer (MIP) based on the molecular recognition theory,which become a potential stationary phase in the solid-phase extraction. The combination of MIP and solid-phase extraction (MISPE) has been used in determining trace analytes in drug, biological and environmental samples. The application of MIP in SPE has become the most promising development of selective SPE method for trace analysis.In this paper,new solid-phase sorbents were synthesized and extraction performances were studied in detail.1. The molecular imprinting of lidocaine (LD) and amiodarone(AD) were studied through computational investigation and template-monomer interaction experiment using UV-vis spectra, and itaconic acid (IA) and methylacrylic acid (MAA) were selected as the best template for LD, 4-vinylpyridine (4-VP) was selected as the best template for AD. The MIP were prepared using ethylene glycol dimethacrylate as cross-linking agent and azobisisobutyronitrile as initiator by bulk polymerization.2. High retention factor and selectivity were achieved by HPLC evaluation of MIP as column sorbent. After optimization of SPE profile, it is determined that the best imprinting effect of the polymer with IA was obtained with loading in acetonitrile, washing with methanol:acetonitrile=5:95,eluting with hydrochloric acid :methanol=5:95, and of the polymer with 4-VP was obtained with loading in pH4.5 buffered solutions,washing with methanol: pH4.5 buffered solutions=30:70, eluting with hydrochloric acid :methanol=5:95.MIP and NIP showed surprisingly high binding capacity in acetonitrile and pH4.5 buffered solutions in breakthrough curve, and their value are 1.62mg and 0.92mg,11.0mg and 2.8mg of drug/100mg of adsorbent, specific adsorption capacity were 0.66mg,8.2 mg drug/100mg of adsorbent.3. Concentration of LD and AD in spiked serum was determined by visable spectrophotometric measurement of the SPE eluent. The experimental results indicated that, LD and AD in serum and injection can be effectively extracted and concentrated by MISPE, and a high degree of clean up and acceptable recoveries was obtained. The developed MISPE procedure allowed the screening of selected arrhythmia drugs in samples. The application of MISPE in pre -treatment of arrhythmia drugs was further developed in the thesis, and provided a new way in sample analysis and detection.
