The Functional Mechanism Of Jinxin Oral Liquid Oppressing RSV Infection The Early Apoptosis Of Cell

Respiratory syncytial virus(RSV),which major causes bronchiolitis and pneumonia, is the leading cause of lower respiratory tract infection in infants.However,the pathogenesis is still unclear. The modern medicine does not have special treatments for the RSV pneumonia. And currently,an RSV vaccine is not yet available.However,the Chinese medicine treatment has certain superiority.Apoptosis can play a significant role in pathogenesis in some virus-infected diseases. Apoptosis is also a common response of cells to infection with RSV. Many viruses have evolved various mechanisms to inhibit the early apoptosis for the replication and spread.JOL has shown in vitro and in viva activity against RSV effeet.However,the precise mechanism of its anti-RSV properties is unknown. It is extrapolated that JOL oppresses RSV by influencing the cell apoptosis. This research is based on the previous study.Objectives:In the present study,we investigated apoptosis during RSV infection in the host cell and expression of Bcl-2 and Bax genes at different points. And we investigated apoptosis during simultaneous incubation of JOL through the observation of the cell apoptosis,as well as expression of Bcl-2 and Bax genes.Methods:With the methods of cell culture,the morphology inspection,medicine serum,RealTime-PCR,Annexin V/PI technique combining with flow cytometry, we detected cell apoptosis,expression of Bcl-2 and Bax genes. The mechanism of JOL against RSV was studied through the above methods. Results were analyzed with statistics software SPSS 13.Conclusions:1) JOL oppresses respiratory syncytial virus.2) It is extrapolated that RSV replication in the cells by suppressing the early cell apoptosis.3) JOL can result in down-regulation of antiapoptotic factor Bcl-2 and up-regulation of pro-apoptotic factors Bax,which reduces the RSV disfusion.
